摘  要:目的探讨急性一氧化碳(CO)中毒后大鼠海马组织基因表达的变化,为发现更稳定、可靠的CO中毒诊断标志物提供线索。方法选取雄性Wistar大鼠20只,均为4月龄。随机分为正常对照组(NC组)10只、急性CO中毒组(ACO-P组)10只。用吸入CO气体法建立急性CO中毒模型,用Biostar R40s基因表达谱芯片检测急性CO中毒14 d后大鼠海马组织基因表达变化的情况。结果急性CO中毒大鼠建模成功,ACO-P组大鼠血液中碳氧血红蛋白(COHb)浓度维持在高水平(>50%);NC组大鼠血中COHb浓度在正常水平(<2%)。急性CO中毒14 d后,大鼠海马组织中共有33条基因出现明显表达变化,其中18条基因上调,15条基因下调;这些基因涉及免疫应答、细胞信号转导、传递蛋白、细胞代谢、衰老等多个方面。结论急性CO中毒后14 d发现多条基因出现差异表达,初步为急性CO中毒的诊断提供了较COHb更为稳定、可靠的生物学标志物。Objective To study the gene expression profile in rat hippocampus after acute carbon monoxide (ACO) poisoning (ACO-P), and provide clues to find more stable and reliable diagnostic biomarkers for ACO-P. Methods Twenty male Wistar rats were randomly divided into normal control(NC) group(n = 10) and ACO-P group(n = 10). ACO-P models were established by inhalation of 99.99 % CO gas. Biostar R40s gene chips were used to examine the change of gene expression in rat hippoeampus at the 14-day after ACO-P. Results Rat ACO-P models were successfully established, and concentration of earboxy-hemoglobin (COHb) in ACO-P group maintained at high level(〉 50 %), while the concentration in NC group was normal(〈 2 %). At the 14--day after ACO-P, 33 genes showed differential expression based on selection criterion, of which 18 genes were up-regulated and 15 genes down-regulated. These genes involved in immunity response, cell signal transduction, transferring, cell metabolism, aging and many other aspects. Conclusion It is demonstrated that several genes differentially expressed at 14 days after ACO-P, which may provide more stable and reliable diagnostic biomarkers for ACO-P than COHb.
