摘 要:利用ISSR分子标记技术,构建了22个甘薯品种的DNA指纹图谱并进行了遗传多样性分析。从40条ISSR引物中筛选了12条引物进行PCR扩增,共扩增出94条带,平均每条引物扩增7.83条带,多态性条带69条,多态性比率为73.40%。将其中3条引物UBC8、UBC35(1)和UBC41结合起来,构建了22个甘薯品种的指纹图谱。聚类分析显示,在阈值为23时,可将22个甘薯品种分为4类,其中,部分品种的ISSR分析与已知系谱相符或与地理分布有一定的相关性。The DNA fingerprinting of 22 sweet potato varieties were constructed using ISSR makers,and genetic diversity was analyzed. From 40 pairs of ISSR primers,12 pairs of primers were selected to amplify,and 94 bands were obtained,on the average,each pair of primer was amplified to 7. 83 bands,69 polymorphic bands were obtained,the polymorphic percentage of bands was 73. 40%. By combining three pairs of primer UBC8,UBC35( 1) and UBC41,22 sweet potato species fingerprinting were constructed.The cluster analysis showed that 22 sweet potato species could be classified into four kinds when threshold was 23,among them ISSR analysis of partial species agreed with pedigree,or related to geography places.
