摘 要:利用S等位基因的4对特异性引物,对284份大白菜高代自交系进行PCR扩增,然后挑选具有单条且目标条带清晰的材料进行克隆、测序、序列分析,初步鉴定大白菜高代自交系的S单元型,并利用亲和指数法、荧光显微检测法验证分子鉴定结果的准确性,这对于加速大白菜育种进程及优势育种高效利用有重要指导意义。结果表明,经PCR扩增获得具有单条且目标条带清晰的材料112份,其包含13个S单元型,分别是S9、S12、S22、S32、S35、S36、S41、S45、S46、S53等10个Ⅰ类S单元型及S40、SD1-8、S44等3个Ⅱ类S单元型。其中,I类S单元型的S46出现频率最高,达到26.79%;其次为II类S单元型的S40,达到24.11%。亲和指数法、荧光显微法鉴定结果基本与分子鉴定结果一致,验证了分子鉴定结果的准确性。相比大白菜类作物中存在的多个S单元型而言,供试材料中S单元型的数目不多,为避免杂交组合配制中出现杂交不结实现象,在进行大白菜遗传育种时,有必要引进和利用更多的S单元型大白菜。The DNA fragments were amplified by PCR with four pairs of specific primers of S allelic genes from all of the 284 Chinese cabbage( Brassica rapa ssp. pekinesis) inbred lines. The lines with single and clear band were selected,of which S haplotypes were preliminarily identified via cloning,DNA sequencing and sequence analysis. The accuracy of molecular identification result was verified by the self-compatibility index method and fluorescence microscopy method. It had guiding significance for speeding up the breeding process and high-efficiency utilization of the advantages of heterosis breeding. The results indicated that 13 S haplotypes were found in 112 materials which had single and clear band,including classⅠhaplotypes,such as S9,S12,S22,S32,S35,S36,S41,S45,S46,S53,and class Ⅱ haplotypes,such as S40,SD1-8 and S44. The highest frequency of S haplotyes was S46 with 26. 79%,and followed by S40 with 24. 11% in this study. The results of the self-compatibility index method and fluorescence microscopy method were consistent with the results of molecular identification,which verified the results of molecular identification. Compared with lots of S haplotypes in Brassica,the S haplotypes in Chinese cabbage inbredlines in this study were fewer,and it was very necessary to introduce and utilize more Chinese cabbage with more S haplotypes.
