AGT基因rs5049位点与饮食饮酒的交互作用对原发性高血压的影响

摘　　要：目的:研究血管紧张素原(angiotensinogen,AGT)rs5049位点的多态性分布与饮食饮酒的交互作用对原发性高血压(essential hypertension,EH)的影响。方法:纳入EH患者400例,正常对照组100例,采用调查表的方式收集饮酒、饮食习惯及相关病史等资料,采用多重高温连接酶技术(Improved Multiple Ligase Detection Reaction,im LDR)检测AGT基因rs5049多态性。运用logistic回归模型分析交互作用对EH发病的影响。结果:AGT基因rs5049位点基因型多态性在EH组与对照组间比较差异具有统计学意义,P<0.05。Logistic回归分析示,AGT基因rs5049位点、饮食、饮酒作为独立影响因素,OR值为1.58、1.92、2.81,P<0.05。三者之间交互作用引起的EH发病OR值为1.46(95%CI:1.08,1.96)。结论 :AGT基因r s5049位点、饮食、饮酒是EH发病的独立影响因素;AGT基因rs5049位点与饮食、饮酒之间存在交互作用,对EH发病有影响。Objective： To invest the relationship between （angiotensinogen AGT）, angiotensin rs5049 loci with drink- ing and eating in patients with essential hypertension （essential hypertension, EH）. Methods： The patients （n=400） with EH were chosen ,another 100 healthy cases were includes as the control group, with the method of questionnaires to collect drinking, eating habits and related history datas, adopting Multiple high temperature Ligase technology （Improved Multiple Ligase Detection Reaction, imLDR） to detect rs5049 AGT gene polymorphism. Using logistic regression model to analyse the effect of interaction on the onset of EH. Results： The comparison difference of rs5049 AGT gene loci genotype poly- morphism between EH group and control group is statistically significant（P〈 0.05）.Logistic regression analysis that, rs5049 AGT gene loci, diet, and alcohol all work as independent factors,each stands for the value of OR by 1.58, 1.58, 1.58, P 〈 0.05）. The interaction between the three of them caused the OR value to be 1.46 （95% CI： 1.08, 1.08）. Conclusion： rs5049 AGT gene loci, diet, and alcohol were all independent factors which are affecting the pathogenesis of EH; The interaction between Rs5049 AGT gene loci ,eating, and drinking are all influential to the onset of EH.
