摘 要:探讨氯卡色林(lorcaserin)对高脂饮食小鼠瘦素及胰岛素信号的影响。30只C57BL/6小鼠随机分为正常组、模型组和给药组;正常组给予标准饲料,模型及给药组给予高脂饲料;同时,给药组每天腹腔注射氯卡色林(10 mg/kg),而其余两组腹腔注射等体积生理盐水;每周测定小鼠体重,直至第12 w取材;试剂盒测定血清胆固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白含量;ELISA法测定血清瘦素和胰岛素水平;q PCR技术检测下丘脑瘦素受体和胰岛素受体基因表达。结果表明,氯卡色林能够有效抑制高脂饲料饮食小鼠体重增加,降低血清胆固醇含量,提高血清胰岛素水平,同时增强下丘脑瘦素受体基因表达。实验结果提示,氯卡色林减肥作用机制与其增强胰岛素和瘦素信号传导有关。The aim of current study was to investigate the effects of lorcaserin on leptin and insulin signaling in mice fed with high fat diet. Thirty mice were randomly divided into three groups, namely normal group, model group and lorcaserin-treated group. Normal group were fed with standard diet ,whereas the other two groups were given high fat diet. Lorcaserin-treated mice were daily intraperitoneally injected with lorcaserin (10 mg/kg). Meanwhile, the other group mice were treated with saline. Body weight of mice was monitored every week and lasted for 12 weeks. Serum cholesterol, triglyceride, high density lipoprotein and low density lipoprotein content were measured with respective kits. Serum leptin and insulin levels were determined by ELISA kits. Gene expression of leptin receptor and insulin receptor in hypothalamus was detected by qPCR assay. The results showed that lorcaserin could effectively inhibit the body weight gain, decrease the content of serum cholesterol, increase the serum insulin level, and enhance the expression of leptin receptor in hypothalamus of high fat diet mice. Therefore, our results suggested that lorcaserin resulted in weight loss through enhancing insulin and leptin signal transduction.
