摘 要:目的:研究小儿清瘟解热颗粒对小鼠免疫功能的影响及体内、外抗菌作用。方法:取Balb/C小鼠70只,将小鼠按体重分为正常组、模型组、玉屏风颗粒组(2.8 g·kg^-1),胸腺肽组(9.2 mg·kg^-1),小儿清瘟解热颗粒低、中、高剂量组(10.92,21.84,43.68 g·kg^-1),每组10只,除正常组外,其余各组通过小鼠ip环磷酰胺构建免疫缺陷小鼠模型,观察小儿清瘟解热颗粒对T,B淋巴细胞增殖的影响及对碳廓清能力,血清、脾细胞溶血素含量的影响;取ICR小鼠只,按体重等级随机分为7组,分别为模型组,阿莫西林组(370 mg·kg^-1),小儿感冒宁合剂组(8.25 m L·kg^-1),小儿清瘟解热颗粒低、中、高剂量组(3.2,6.4,12.8 g·kg^-1),每组20~22只,采用小鼠腹腔注射金黄色葡萄球菌构建体内细菌感染模型,观察动物死亡数、死亡率,药物的保护率,动物的平均存活天数及生命延长率;在体外采用试管法观察药物对金黄色葡萄球菌、表皮葡萄球菌、白色葡萄球菌、大肠埃希菌、绿脓杆菌、乙性溶血性链菌、白色念珠菌、淋球菌及变形杆菌的最低抑菌浓度(MIC)。结果:小儿清瘟解热颗粒低、中剂量均可显著提高脂多糖(LPS),刀豆球蛋白A(Con A)诱导的淋巴细胞增殖;对于金黄色葡萄球菌体内感染模型,小儿清瘟解热颗粒可使动物死亡率明显降低,动物存活天数明显延长(与模型组比较P〈0.05,P〈0.01);同时,小儿清瘟解热颗粒体外对金黄色葡萄球菌、表皮葡萄球菌、白色葡萄球菌、大肠埃希菌、绿脓杆菌、乙性溶血性链菌、白色念珠菌及变形杆菌生长均有不同程度的抑制作用。结论:小儿清瘟解热颗粒对免疫低下小鼠的免疫功能具有一定的提高作用,在体内对金黄色葡萄球菌感染致小鼠死亡具有明显的保护作用,在体外对多种细菌的生长具有不同程度的抑制作用。Objective: To study the effect of Xiaoer Qingwen Jiere Granule in regulating immune function in mice,and its antibacterial efficacy in vitro or in vivo. Method: Totally 70 Balb / C mice were divided into following groups: normal,model,Yupingfeng granule,thymosin,and low,medium,high doses of Xiaoer Qingwen Jiere granules( 10. 92,21. 84,43. 68 g crude drug / kg) according to body weight,with 10 in each group. Immune deficiency model was constructed through intraperitoneal injection with cyclophosphamide in all groups except for the normal group,in order to observe the influence of Xiaoer Qingwen Jiere granule on the T or B lymphocyte proliferation,carbon clearance ability and serum and hemolysin content. ICR mice were randomly divided into seven groups by weight: model group,amoxicillin group( 370 mg ·kg- 1),Xiaoer Ganmaoning mixture group( 8. 25 m L·kg- 1) and low,medium,high-dose Xiaoer Qingwen Jiere granules groups,with 20 to 22 mice in each group. Bacterial infection model was constructed through intraperitoneal injection with Staphylococcus aureus to observe the animal mortality rate,drug protection and the average survival days of animals. The minimal inhibitory concentrations( MIC) of S. aureus,S. epidermidis,Escherichia coli,Candida albicans were observed by using the tube method Qingwen Jiere. Result: Low and medium doses of Xiaoer Qingwen Jiere granules can increase the lymphocyte proliferation induced by lipopolysaccharide( LPS) and Concanavalin( Con A)significantly. In the model induced by staphylococcus aureus,Xiaoer Qingwen Jiere granules can reduce the mortality significantly and prolong the survival time and inhibite S. aureus( P〈0. 05,P〈0. 01),S. epidermidis,E. coli,C. albicans to varying degrees. Conclusion: Xiaoer Qingwen Jiere granule can improve the immune function of immune compromised mice,protect the mice infected by staphylococcus aureus from death in vivo,and inhibit the growth of a variety of bacteria inordinately.
