摘 要:以7-苯乙酰胺基-3-氯甲基头孢烷酸对甲氧基苄酯(GCLE)为原料合成了7-苯乙酰氨基-3-(丙-1-烯基)-3-头孢烯-4-羧酸-4-甲氧基苄酯(GPRE),并对Wittig反应进行了研究。结果标明:氯化锂存在时,在v(二氯甲烷/m L)∶v(N,N-二甲基乙酰胺/m L)∶w(Wittig试剂/g)=3∶2∶1、反应温度-10℃的条件下,GPRE Z、E比值超过13.5,主峰(Z+E)超过97.0%。This paper reported the Wittig reaction using (6R-trans)-5-Thia-l-azabicyclo [4.2.0]oct-2-ene-2- carboxylic acid, 3-chloromethyl-8-oxo-7-[(phenylacetyl)amino]-diphenylmethyl ester(GCLE) as the raw material for the synthesis of 4-methoxybenzy1-7-phenylacetamido-3-(propen-1-alkenyl)-3-cephem-4-carboxylate (GPRE). The results indicated that the Z/E value of GPRE was greater than 13.5, and the Z+E value is greater than 97.0% under the following reaction conditions: Lithium chloride was used as the catalyst; the ratio of dichloromethane to N,N-dimethylacetamide (both in volume) and to Wittig reagent (weight) was 3:2:1; the reaction temperature was of -10℃.
