摘  要:目的研究活性维生素D3与特异性脱敏疗法治疗小鼠过敏性鼻炎疗效的差异性。方法 2014年7—11月,选取SPF级雌性BALB/c小鼠32只为研究对象。制备葎草花粉粗提液,克隆表达p TSX2,十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)检测葎草花粉粗提液蛋白组分和p TSX2活性。采用简单随机抽样法将32只小鼠分为4组,分别为磷酸盐缓冲液(PBS)空白对照组、模型组、p TSX2脱敏治疗组、活性维生素D3治疗组,每组8只。对各组小鼠进行过敏性鼻炎造模并给予不同的治疗。通过比较小鼠鼻部症状及行为学评分、气道高反应性、血清特异性s Ig E、s Ig G2a水平、脾脏细胞培养液上清液中细胞因子〔白介素4(IL-4)、干扰素γ(IFN-γ)〕水平、鼻黏膜病理改变来评估两种疗法的差异。结果模型组、p TSX2脱敏治疗组、活性维生素D3治疗组小鼠鼻痒、喷嚏、鼻部分泌物、行为、总分评分高于PBS空白对照组(P<0.05);p TSX2脱敏治疗组、活性维生素D3治疗组小鼠鼻痒、鼻部分泌物、行为、总分评分低于模型组(P<0.05);活性维生素D3治疗组小鼠喷嚏评分高于模型组(P<0.05);活性维生素D3治疗组小鼠总分评分高于p TSX2脱敏治疗组(P<0.05)。乙酰甲胆碱浓度为0.00、6.25、12.50、25.00、50.00、100.00 mg/ml时,模型组增强呼气间歇(Penh)值与PBS空白对照组比较,差异有统计学意义(P<0.05);乙酰甲胆碱浓度为50.00、100.00 mg/ml时,p TSX2脱敏治疗组、活性维生素D3治疗组Penh值与PBS空白对照组比较,差异有统计学意义(P<0.05);乙酰甲胆碱浓度为25.00、50.00、100.00 mg/ml时,p TSX2脱敏治疗组、活性维生素D3治疗组Penh值与模型组比较,差异有统计学意义(P<0.05)。模型组、p TSX2脱敏治疗组、活性维生素D3治疗组血清中s Ig E、s Ig G2a及脾脏细胞培养液上清液中IL-4、INF-γ水平高于PBS空白对照组(P<0.05);p TSX2脱敏治疗组、活性维生素D3治疗组s Ig E、IL-4水平Objective To investigate the differences between 1,25( OH)2D3and specificity immunotherapy therapy on mice with allergic rhinitis. Methods From July to December in 2014,32 SPF female BALB / c mice were selected as research subjects. The crude extract of humulus scandens pollen was prepared, and cloned expression was made for p TSX2 protein. SDS-PAGE was used to test the protein components and p TSX2 protein activity of the crude extract of humulus scandens pollen. Using simple random sampling method,the 32 mice were divided into 4 groups: PBS blank control group,model group,p TSX2 immunotherapy group and 1,25( OH)2D3group. Each group had 8 mice and was administrated with different treatment methods. By the comparison of nasal symptoms,behavioral scores,airway hyperresponsiveness,serum s Ig E and s Ig G2 a levels,IL-4 level, IFN-γ level and the pathological changes of nasal mucosa, the differences between the two therapies were evaluated. Results The model group,the p TSX2 immunotherapy group and the 1,25( OH)2D3group were higher than the PBS blank control group in the scores of rhinocnesmus,sneeze,nasal secreta,behavior and total score( P〈0. 05); the p TSX2 immunotherapy group and the 1,25( OH)2D3group were lower than the model group in the scores of rhinocnesmus, nasal secreta,behavior and total score( P〈0. 05); the 1,25( OH)2D3group was higher than the model group in the score of sneeze( P〈0. 05); the 1,25( OH)2D3group was higher than the p TSX2 immunotherapy group in total score( P〈0. 05).When the alkali concentration of methacholine was 0. 00,6. 25,12. 50,25. 00,50. 00 and 100. 00 mg / ml,the Penh value of the model group was significantly different from the PBS blank control group( P〈0. 05); when the alkali concentration of methacholine was 50. 00 and 100. 00 mg / ml,the p TSX2 immunotherapy group and the 1,25( OH)2D3group were significantly different from the PBS blank control group in Penh value( P〈0. 05); when the alkali concentration of m
