摘 要:目的:运用代谢组学技术探究脾气虚模型大鼠血清小分子代谢谱群特征变化,从而探索脾气虚证的生物学本质。方法:14只SPF级雄性SD大鼠按随机数字表法分为正常对照组和脾气虚证模型组各7只,模拟中医病因建立脾气虚大鼠模型。运用LC-Q/TOF-MS质谱技术对2组大鼠血清样本进行检测,通过主成分分析(PCA)、最小二乘判别分析(PLS-DA)和正交偏最小二乘分析(OPLS-DA)比较2组大鼠血清代谢图谱,寻找差异性表达代谢物。结果:脾气虚大鼠与正常大鼠的血清代谢图谱间存在显著差异。与正常大鼠比较,脾气虚大鼠血清中葡萄糖、琥珀酸等小分子代谢物含量显著上升,雄酮、亚油酸等含量显著下降,通过OPLS-DA分析得到一些脾气虚证的潜在生物标志物。结论:脾气虚大鼠呈现出不同于健康大鼠的血清代谢谱群特征,该证型存在多种代谢通路异常,可能伴随机体能量供应障碍,也可能诱发糖尿病及心血管疾病。Objective: To study the changes in the characteristics of serum metabolism in rats with spleen-qi deficiency, in order to investigate the essence of spleen-qi deficiency. Methods: 14 male SD SPF rats were randomly assigned into two groups which were the spleen-qi deficiency model group and the normal control group. Each group had 7 rats. Spleen-qi deficiency rat model was established simulating traditional Chinese medicine cause of disease. LC-Q/TOF- MS was used to exam the serum samples of the rats. The serum metabolism was compared between the spleen-qi deficiency model group and the normal control group via PCA, PLS-DA and OPLS-DA analysis. Metabolites with levels significantly different between these two groups were identified. Results: There were significant differences in serum metabolism between rats in the spleen-qi deficiency group and those in the normal control group. Levels of some metabolites such as glucose, succinic acid. were significantly increased and those of androsterone, linoleic acid. were significantly decreased in rats with spleen-qi deficiency compared to healthy rats. Conclusion : Rats with spleen-ql deficiency showed different characteristics of serum metabolism compared to healthy rats. Spleen-qi deficiency was accompanied with abnormality in many metabolism systems. It was also associated to insufficient energy supply and may lead to diabetes and some cardiovascular diseases.
