摘 要:目的:分析活血化痰方干预糖尿病大鼠的尿液代谢物变化规律。方法:采用高脂饮食联合链脲佐菌素制备糖尿病大鼠,设立模型组、对照组、中药组和西药组。第6周末检测各组大鼠血糖、血脂,并运用气相色谱-质谱联用法检测第3、6周尿液,对代谢物进行主成分分析。结果:大鼠成模后判别属脾气亏虚、痰瘀互阻证,与对照组比较,模型组血糖、血脂升高,第6周抗坏血酸、葡萄糖酸、丙氨酸、十八烷酸、戊二酸含量增加;模型组第6周较第3周上述代谢物含量增加;中药组第6周较第3周抗坏血酸含量下降。结论:尿液代谢物变化提示活血化痰方可以改善糖尿病大鼠体内氧化应激状态及脂类代谢异常。Objective:To explore the characteristics of urine metabolites in diabetic rats treated by the compound of invigorating blood and reducing phlegm. Methods: Diabetic rats were duplicated with high-fat diet and injection of streptozotocin. There were four groups: the model group, the control group, the compound group and the western medicine group. Blood glucose and lipid were measured in the 6th week after duplicating and gas chromatography-mass spectrometry (GC-MS) was applied to measure the urine samples in the 3 rd and 6th week. The principal component analysis (PCA)was used to calculate the main metabolites. Results:The syndrome of spleen-deficiency and retention of phlegm-blood stasis was confirmed after duplication. Compared with the control group, the blood glucose and low density lipoprotein in model group increased significantly; L-Ascorbic acid, D-Gluconic acid, α-Alanine, Octadecanoic acid, Pentanedioic acid increased in the 6th week. Compared with the same group in the 3rd week, the above metabolites increased in the model group in the 6th week,and the L-Ascorbic acid decreased in the compound group in the 6th week. Conclusion: The changes of urine metabolites uncovered the compound of invigorating blood and reducing phlegm can improve the disorder of oxidative stress and lipid metabolism in diabetic rats.
