清肝化瘀方含药血清对人巨细胞病毒感染后人胚肺成纤维细胞细胞病变及增殖的影响

摘　　要：目的探讨清肝化瘀方含药血清对人巨细胞病毒（HCMV）感染后人胚肺成纤维细胞（HELF）细胞病变及增殖活性的影响。方法以体外培养的HELF为靶细胞,将清肝化瘀方含药血清和更昔洛韦（GCV）含药血清分别作用于HCMV AD169株感染的HELF,采用倒置相差显微镜动态观察细胞病变,MTT检测细胞增殖活性。结果清肝化瘀方含药血清、GCV含药血清最大无毒浓度均为血清稀释度10-3。HCMV感染后24~72 h、168 h细胞病变（＋）;24~72 h、144 h增殖活性减低。清肝化瘀方含药血清、GCV含药血清干预后,细胞病变均消失;干预24~96 h增殖活性均逐渐升高,与病毒组比较,均以72 h时升高为主,差异有统计学意义（P〈0.05）。2组间细胞病变及增殖活性比较,差异均无统计学意义（P〉0.05）。结论清肝化瘀方、GCV含药血清均可通过遏制HCMV感染后细胞病变,调节HELF增殖而发挥抗病毒作用。Objective To investigate Qinggan Huayu Prescription medicated serum on the lesion and proliferation activity of human embryo fibroblast （HELF） after human cytomegalovirus （HCMV） infection. Methods With HELF cultivated in vitro as target cells, Qinggan Huayu Prescription medicated serum and ganciclovir （GCV） contained serum were used to act on HCMV AD169 infected HELF, the inverted phase contrast microscopy were used to observe the cell lesions dynamically, and MTT to detect the cell proliferation activity. Results The maximum non-toxic concentration of Qinggan Huayu Prescription contained serum and serum containing GCV were both serum dilution 103. At 24-72 h, 168 h cell lesions （ ＋ ） , at 24-72 h after HCMV infection, 144 h, the proliferation activity was decreased. After intervention with Qinggan Huayu Prescription medicated serum and GCV contained serum, cell lesions disappeared; with intervention at 24-96 h, the proliferation activity were increased, compared with the virus group, they were increased mainly at 72 h, the difference was statistically significant （P 〈 0. 05 ）. The difference in cell lesion and proliferation activity between two groups had no statistical significance （P 〉 0. 05 ）. Conclusion Qinggan Huayu Prescription and GCV can inhibit cell lesion after HCMV infection and regulate HELF proliferation so as to produce antiviral effects.
