摘 要:目的:研究不同生长期穿心莲叶中6种同工酶的酶谱变化规律,为探讨同工酶与穿心莲次生代谢产物形成之间的关系奠定基础。方法:以不同生长时期的穿心莲顶端全展叶为供试材料,采用聚丙烯酰胺凝胶电泳(PAGE)法得到穿心莲叶过氧化物酶(POD)、超氧化物歧化酶(SOD)、多酚氧化酶(PPO)、过氧化氢酶(CAT)、细胞色素氧化酶(CYT)及酯酶(EST)同工酶的电泳酶谱,对不同生长期穿心莲叶中上述6种同工酶图谱进行比较分析。结果:不同生长期穿心莲叶中6种同工酶的酶带亮度(酶活性)均随着生长时期的不同显示出明显差别,且均具有各自的主要特征酶带。POD、CYT和PPO同工酶酶谱在质(酶带数)和量(酶活性)上均表现出不同程度的差异,其酶活性在花果期显著增强,其中POD受物候期的影响最为显著;EST同工酶的酶活性也在花果期明显提高;CAT和SOD同工酶在穿心莲生长过程中相对稳定。结论:POD、EST、PPO以及CYT同工酶酶谱的变化可以明显反映不同生长期穿心莲的生长发育特异性,尤其是POD可以作为观察穿心莲生长变化以及深入研究次生代谢产物生成的一种生理指标。Objective: The influence of phonologicalstages (vegetative, squaring, flowering and fruiting stages) on six kinds of isozymes of Andrographis paniculata leaf were investigated, which may offer references forunderstanding the relationshi Pbetween the isozymes and generation of secondary metabolites. Methods: The first fully developed leaves from the to Pwere used as thetested materials. The eleetrophoresis spectrums of six isoenzymes including peroxidase (POD) , superoxide dismutase (SOD) , Polyphenol oxidase ( PPO ) , Catalase (CAT) , Cytochrome Oxidase (CYT) and esterase (EST) were obtained by polyacrylamide gel electrophoresis (PAGE) method and then analyzed consequently. Results: The aforementioned isozymes showed significant difference with different phonological stages. The number of POD, EST, CYT and PPO enzyme band and their band intensity increased at flowering and fruiting period, around September and October. POD was affected most significantly. While CAT and SOD werestable during the growth period. Conclusion: Thechanges of POD, EST, CYT and PPO isoenzyme pattern, especiallythe POD can clearly reflect the growth and development specificity of A. paniculata at different growth period and itmay be used as aphysiological and chemical index for the further study of A. paniculata.
