摘  要:目的:研究四倍体金银花的体外抗菌和抗病毒作用。方法:以二倍体金银花为对照,用最小抑菌浓度(MIC)和最小杀菌浓度法(MBC)测定四倍体金银花水提物对金黄色葡萄球菌、乙型链球菌、大肠杆菌、绿脓杆菌、白色念球菌、肺炎克雷伯菌的抗菌和杀菌作用。采用细胞病变效应法(CPE)观察四倍体金银花水提物对狗肾传代细胞(MDCK)的最大无毒浓度(TC0)和半数中毒浓度(TC50)。将100TCID50的甲型流感病毒FM1加入MDCK细胞培养板中,吸附1 h后弃病毒液于MDCK细胞培养板中加入四倍体金银花水提液的最大无毒浓度稀释液,通过CPE法观察提取物对病毒致细胞病变作用的影响。结果:四倍体金银花水提物对金黄色葡萄球菌、乙型链球菌、大肠杆菌、绿脓杆菌、白色念球菌、肺炎克雷伯菌均具有抑菌和杀菌作用,其中对金黄色葡萄球菌的抑菌和杀菌作用最为明显,四倍体对乙型链球菌抑菌强度高于二倍体,对肺炎克雷伯菌的杀菌强度高于二倍体。四倍体金银花水提物在浓度为0.030 5 mg·m L-1对甲型流感病毒FM1所致的细胞病变作用显示抑制作用,而其对照药材二倍体金银花水提物在浓度为0.488 0、0.244 0 mg·m L-1对流感病毒所致的细胞病变作用显示抑制作用。结论:四倍体金银花具有体外抗菌及抗流感病毒的作用。Objective: To study the antimicrobial and antiviral activities of the tetraploid Lonicerae Japonicae Flos in vitro. Methods : Compared with the diploid Lonicerae Japonicae Flos, the minimum inhibitory concentration ( MIC ) and the minimum bactericidal concentration ( MBC ) were used to evaluate the antimicrobial effects of the aqueous extract of the tetraploid Lonicerae Japonicae Flos against Staphylococcus aureus , Beta hemolytic streptococcus, Escherichia coli , Pseudomonas aeruginosa, Candida albicans and Klebsiella pneumonia. The maximum non-toxic concentration (TC0 ) and median toxic concentration ( TCs0 ) of the aqueous extract of the tetraploid on Madin-Dar by Canine Kidney (MDCK) cell were detected by cytopathic effect (CPE). Adding 100TCID50 of influenza A virus FM1 into cell microtiter plates, then getting rid of the all the virus solutions after adsorbed in the plates for 1 hour, adding the maximal non-toxic concentration dilution. The influences of the aqueous extract on the CPE of FM1 were observed by using CPE method. Results: The aqueous extract of the tetraploid Lonicerae Japonicae Flos had inhibitory effect and bactericidal action against S. aureus, B. streptococcus, E. coli, P. aeruginosa, C. albicans and K. pneumonia. It had strong inhibitory effect and bactericidal action against S. aureus among the six strains. The aqueous extract of the tetraploid had stronger inhibitory effect against B. streptococcus and stronger bactericidal action against K. pneumonia than that of the diploid. The aqueous extract from the tetraploid showed inhibitory effect on the CPE induced by FMlwhen the concentration of the aqueous extract was 0. 0305 mg· mL-1. While its reference herb, the diploid Lonicerae Japonicae Flos, showed inhibitory effect on the CPE when the concentration of the aqueous extract was 0. 488 0 and 0. 244 0 mg· mL-1. Conclusion: The tetraploid Lonicerae Japonicae Flos had the antimicrobial and antibacterial activity in vitro.
