摘  要:目的:研究丹参总酚酸、总丹参酮对酪氨酸酶的抑制作用机理。方法:采用紫外-可见分光光度法测定490nm处酪氨酸酶催化产生的多巴醌的吸光度,并通过Lineweaver-Burk双倒数作图法计算酪氨酸酶的Km和Vm值,考察丹参总酚酸、总丹参酮对酪氨酸酶的抑制作用及作用机理。结果:酪氨酸酶的Km=0.2067mg·ml-1,Vm=0.0222·min-1,总丹参酮在浓度为2mg·ml-1抑制率为0.00%,丹参总酚酸浓度达到10mg·ml-1后Km变大,Vm变小。结论:总丹参酮对酪氨酸酶无抑制作用;丹参总酚酸对酪氨酸酶具有可逆抑制作用,抑制类型属于混合I型。Objective To study the inhibition of Salvia miltiorrhiza total phenolic acid,Total tanshinone on tyrosinase and their mechanism. Methods Using ultraviolet-visible spectrophotometry determines the absorbance of tyrosine enzyme catalysis to produce Dopaquinone at 490nm and calculates the Kin, Vm values of tyrosinase through the method of Lineweaver-Burk Plot;Studying the inhibition of Salvia miltiorrhiza total phenolic acid,Total tanshinone on tyrosinase and their mechanism. Results Tyrosinase Km=0.2067mg. ml-l,Vrn= 0.0222~ min-l,when the concentration of Total tanshinone reaching 2mg. ml-1,the inhibition rate was 0.00% ;when the concentration of Salvia miltiorrhiza total phenolic acids reaching 10mg^ml-1;the higher Km become,the lower V~ become. Conclusion Total tanshinone had no effects on tyrosinase,the inhibition of Salvia miltiorrhiza total phenolic acid on tyrosinase was reversible,the Inhibit type belongs to mixed type I.
