摘　　要：目的观察脐带间充质干细胞(Wharton's jelly mesenchymal stem cells,WJ-MSCs)移植对异丙基肾上腺素(ISO)致心力衰竭(Heart failure,HF)大鼠炎性细胞因子表达的影响。方法雄性Wistar大鼠50只,随机分为空白组、模型组和干细胞移植组。空白组不作处理正常饲养;模型组和干细胞移植组予200 mg/kg多点皮下注射异丙肾上腺素,隔24h注射1次共2次,制造心力衰竭大鼠模型成功后1周,干细胞移植组心肌内注射1 m L(1×106MSCs)干细胞悬液;模型组予心肌内注射等量生理盐水。移植前、移植后4周分别用酶联吸附免疫反应法(ELISA)测定三组大鼠血清中N末端脑利钠肽前体(NT-pro BNP)、超敏C反应蛋白(hs-CRP)及肿瘤坏死因子(TNF-α)水平,并取大鼠心脏组织行免疫组化染色。结果移植前,心力衰竭模型组和干细胞移植组大鼠NT-pro BNP、hs-CRP和TNF-α较空白组大鼠增高显著(P<0.05);移植后4周,干细胞移植组NT-pro BNP、hs-CRP和TNF-α与模型组比较,明显降低,差异具有统计学意义(P<0.05)。结论脐带间充质干细胞移植治疗可显著下调心力衰竭大鼠心脏hs-CRP和TNF-α炎性细胞因子表达,改善心衰症状。Objective To explore the effect of transplanted Wharton＇s jelly mesenchymal stem cells（WJ-MSCs）on theexpression of inflammatory cytokines in the rats with heart failure. Methods A total of 50 Wistar rats were randomly dividedinto three groups： control group, model group and transplantation group. Heart failure was induced in model group andtransplantation group by isoproterenol（ISO）injection. Rats in control group were untreated and normally bred; One week afterthe heart failure established,1×106MSCs（1 m L）were transplanted to the rats by intramyocardial injection in transplantationgroup. Rats in model group were treated with the same dose of NS.Before and 4 weeks after cell transplantation, ELISA wereperformed to investigate the expression of NT-pro BNP,hs-CRP and TNF-α, and the rats were sacrificed for immunochemicalanalysis. Results Before transplantation,levels of NT- pro BNP,hs- CRP and TNF- α in patients of model group andtransplantation group were higher than those in control group.However,four weeks after cell transplantation, the expression ofTNF- α,hs- CRP,and NT- pro BNP in the transplantation group were much lower than those in the model group（P0.05）.Conclusion WJ-MSCs transplantation can improve cardic function and down-regulate the expression of TNF-α, hs-CRPand NT-pro BNP in a rat with heart failure induced by ISO.
