摘  要:为了建立八棱海棠的组织培养快繁体系,以八棱海棠单芽茎段为外植体,以MS为基本培养基进行组织培养研究。结果表明,5月为单芽茎段外植体获取的最佳时间,此时污染率低,死亡率低;最佳丛生芽诱导培养基为MS+6-BA 1.0 mg/L+NAA 0.1 mg/L,诱导率为53.2%;MS+6-BA 2.0 mg/L+NAA 0.2 mg/L为最佳增殖培养基,暗培养2周,增殖系数为4.73;生根培养以1/2 MS+IBA 0.3 mg/L+NAA 0.2 mg/L较好,平均根长为5.2 cm,生根率为90.0%。In order to establish tissue culture rapid propagation for Malus robusta, relevant study was made by tak-ing single-bud stalk segment as explant, and MS as the basic medium. The result shows that the May is the besttime for obtaining explant, and both contamination rate and death rate are lowest at that time. MS+6-BA 1.0mg/L+NAA 0. lmg/L is the best culture medium for inducing, and the rate is 53.2 %. MS+6-BA 2. 0 mg/L+NAA0. 2 mg/L is the best culture medium for multiplication, and the coefficient is 4. 73 with two weeks dark culture.1/2 MS+IBA 0. 3 mg/L+NAA 0.2 mg/L is appriopriate for rooting culture , and the average root length is 5.2em, while rooting rate is 90. 0 %.
