摘  要:目的探究补骨脂乙素(Isobavachalcone,IBC)对人舌鳞状细胞癌Tca8113细胞迁移和侵袭的抑制作用及其可能的作用机制。方法将不同浓度的IBC作用于体外培养的Tca8113细胞,通过MTT法检测细胞增殖抑制率;划痕实验和Transwell实验分别检测细胞迁移和侵袭能力;Western blot法检测Akt、p-Akt、MMP-2和MMP-9蛋白的表达情况。结果 IBC以浓度和时间依赖的方式有效抑制Tca8113细胞增殖。IBC降低Tca8113细胞迁移及侵袭能力。Western blot法显示IBC能够以浓度相关性下调p-Akt、MMP-2和MMP-9蛋白的表达,而Akt蛋白水平不受IBC处理剂量的影响。结论 IBC可抑制Tca8113细胞的增殖,迁移和侵袭,其作用与下调MMP-2和MMP-9蛋白及抑制其上游Akt的磷酸化有关。Aim To explore the inhibitory effect of isobavachalcone( IBC) on migration and invasion of tongue squamous cell carcinoma Tca8113 cells and its possible mechanism. Methods Tca8113 cells were treated in different concentrations of IBC in vitro. Cell proliferation was detected by MTT; Wound healing assay and Transwell chamber assay were used to detect the ability of cell migration and invasion; Western blot was applied to detect the expression of Akt,p-Akt,MMP-2 and MMP-9 proteins. Results IBC could inhibit the proliferation of Tca8113 cells in a concentration-and time-dependent manner. IBC can reduce cell migration and invasion. Western blot showed that IBC could an decrease the expression of p-Akt,MMP-2 and MMP-9 proteins in a concentration-dependent manner.However,the level of Akt was not affected by the concentration of IBC treatment. Conclusion IBC could inhibit the proliferation, migration and invasion in Tca8113 cells and its mechanism may be associated with the down-regulation of MMP-2 and MMP-9 proteins and the inhibition of phosphorylation of upstream Akt.
