摘 要:目的探讨肾脏损伤因子(KIM-1)基因2个SNP位点与中国山东汉族人群糖尿病肾病(DKD)的相关性,为糖尿病肾病的早期预防提供理论依据。方法收集2型糖尿病肾病(DKD)患者274例,健康对照(NC)个体248例。提取以上样品的全血基因组DNA,用聚合酶链式反应—限制性酶切片段长度(PCR-RFLP)多态性技术进行检测,对KIM-1基因启动子区的-416G>C和-1454G>A两个SNP位点的基因型频率、等位基因频率进行比较和分析。结果健康对照组与DKD组的-416G>C基因频率和基因型频率分布的差异有统计学意义(P=0.004,P=0.001)。健康对照组与DKD组的-1454G>A基因频率和基因型频率没有统计学差异。结论 -416G>C位点的多态性与中国汉族人群DKD的发生有关,GG基因型可能增加DKD的发病风险。Objective To explore the effect of KIM- 1 polymorphism in diabetic kidney disease (DKD). Methods Two KIM-1 gene polymorphism had been analyzed in 274 DKD patients and 248 normal controls by polymerase chain reaction-restriction fragment length polymorphism. Results There was significantly difference on the distribution of KIM-1 -416G 〉 C polymorphism and the allele frequency between DKD and normal control group (P 〈0.01 ), but without significant difference on KIM-1 - 1454G 〉 A between them. Conclusion The distribution of KIM-1 gene-416G 〉 C polymorphism is associated with DKD. The GG genotype of the-416G 〉 C polymorphism of K/M- 1 gene is a genetic risk factor for diabetic kidney disease.
