祁州漏芦对肝衰竭小鼠肝细胞凋亡的拮抗作用

摘　　要：目的研究祁州漏芦提取物(RUE)对内毒素和半乳糖胺诱发的急性肝衰竭小鼠肝细胞凋亡的拮抗作用。方法应用内毒素和半乳糖胺制备小鼠急性肝衰竭模型,采用分光光度法检测血清谷丙转氨酶(ALT)和谷草转氨酶(AST),用酶联免疫吸附(ELISA)法检测血清肿瘤坏死因子-α(TNF-α),用苏木素-伊红(HE)染色法观察肝组织病理学变化,用比色法检测肝脏Caspase-3、Caspase-8和Caspase-9活化水平,用琼脂糖凝胶电泳法检测小鼠肝细胞DNA断裂情况。结果与对照组比较,模型组小鼠血清ALT、AST活性升高,肝组织病理学损伤加重;血清TNF-α增高;肝组织Caspase-3、8和9相对活性升高,肝细胞DNA断裂程度增高。与模型组比较,RUE组小鼠血清ALT、AST活性降低,肝组织损伤减轻;血清TNF-α水平下降;肝Caspase-3、8和9相对活性降低,肝细胞DNA断裂程度减少。结论祁州漏芦对内毒素和半乳糖胺致急性肝衰竭小鼠肝细胞凋亡具有拮抗作用。Objective To investigate the antagonistic effect of Rhaponticum uniflorum extract （RUE） on hepatocyte apoptosis of mice with acute hepatic failure induced by endotoxin and D - galactosamine. Methods Endotoxin and galactosamine were given in- traperitoneally to the mice of groups, then the pathological changes was detected with hematoxylin ＆ eosin （HE） staining, tumor necrosis factor-α （TNF -α） was detected by ELISA method, the activities of alanine aminotransferase （ALT）, aspartate amin- otransferase （AST） , Caspase- 3, Caspase -8 and Caspase- 9 were detected by the colorimetric method, and DNA damage was analyzed using the electrophoretic method. Results Compared with the control group, the serum ALT and AST activities increased, hepatic lesions worsened, serum TNF - α content was elevated, the relative activities of hepatic Caspase - 3,8 and 9 increased, and DNA laddering of liver cells increased in the model group. Compared with the model group, serum ALT and AST reduced, the he- patic lesions alleviated, serum TNF - α and relative activities of hepatic Caspase - 3,8 and 9 reduced, and DNA laddering of liver cells decreased in 240 mg/kg RUE group. Conclusion Rhaponticum uniflorum has an antagonistic effect on hepatocyte apoptosis of mice with acute hepatic failure induced by endotoxin and D - galactosamine.
