摘　　要：目的:研究紫杉醇联合顺铂对甲状腺癌细胞SW579增殖、迁移和侵袭作用的影响及其作用机制。方法:将细胞分为空白对照组、紫杉醇(3μmol/L)组、顺铂(30μmol/L)组和联合用药(紫杉醇3μmol/L+顺铂30μmol/L)组,培养48 h。采用MTT法测定相对细胞活力;流式细胞术测定细胞周期;Transwell法检测细胞迁移和侵袭能力;Western blot法检测细胞中人第10号染色体缺失的磷酸酶及张力蛋白同源的基因(PTEN)、蛋白激酶B(AKT)、细胞周期蛋白D1(Cyclin D1)、p27、基质金属蛋白酶(MMP)-2、MMP-9的表达。结果:与空白对照组比较,各给药组细胞的相对细胞活力降低,细胞发生G1期阻滞,细胞的迁移和侵袭能力降低,细胞中PTEN、p27表达增强,AKT磷酸化及Cyclin D1、MMP-2和MMP-9表达减弱,差异均有统计学意义(P<0.05);与单用药组比较,联合用药组作用效果增强,以上指标差异均有统计学意义(P<0.05)。结论:紫杉醇和顺铂联用后对甲状腺癌细胞SW579增殖、迁移和侵袭的抑制作用增强,其机制可能与上调PTEN、p27的表达,下调Cyclin D1、MMP-2、MMP-9的表达及抑制AKT磷酸化有关。OBJECTIVE：To study the effects of paclitaxel combined with cisplatin on the proliferation,migration and invasion of thyroid cancer cells SW579 and its mechanism. METHODS：Cells were divided into blank control group,paclitaxel group（3μmol/L）,cisplatin group（30 μmol/L）,drug combination group（paclitaxel 3 μmol/L＋cisplatin 30 μmol/L）. 48 h after culture,the relative cell activity was measured by MTT assay. Cell cycle was detected by flow cytometry. Migration and invasion of cell was tested by Transwell assay. The expression of phosphatase and tensin homolog deleted on chromosome ten（PTEN）,protein kinase B（AKT）,Cyclin D1,p27,matrix metalloproteinase（MMP）-2 and MMP-9 were detected by Western blot. RESULTS：Compared with blank control group,relative cell activity of all treatment groups were decreased;paclitaxel or plus cisplatin also made cell cycle arrest in G1 phase,and migration and invasion ability of cell were decreased;the expression of PTEN and p27 remarkably increased,while the expression of Cyclin D1,MMP-2,MMP-9 and phosphorylation of AKT were obviously reduced,with statistical significance（P〈0.05）. Compared with single drug group,the effect of drug combination group strengthened,with statistical significance in above indicators（P〈0.05）. CONCLUSIONS：The inhibition effect of paclitaxel combined with cisplatin on the proliferation,migration and invasion of thyroid cancer cells SW579 cell will be strengthened,by a mechanism of up-regulating the expression of PTEN and p27,down-regulating the expression of Cyclin D1,MMP-2 and MMP-9,inhibiting phosphorylation of AKT.
