摘 要:为了研究热应激对猪卵母细胞成熟的影响,以及热应激过程中培养基添加褪黑素是否能够缓解生理过高热对卵母细胞的损伤,试验对体外成熟过程中(0~2h)的卵母细胞进行热应激(41℃),并在成熟培养液(M199)中添加褪黑素(1×10-9mol/L),对其后期发育和囊胚细胞数、囊胚细胞凋亡进行检测。结果表明:热应激能够显著降低猪卵母细胞的囊胚率和囊胚细胞数,囊胚细胞凋亡率显著升高,而添加褪黑素后能够缓解热应激对卵母细胞的影响。说明热应激能够影响卵母细胞的成熟,并对卵母细胞的后期发育产生影响,同时在培养液中加入浓度为1×10-9mol/L的褪黑素能够缓解热应激损伤。To study the effect of heat stress on the maturation of procine ooeytes, and whether adding melatonin to the culture medium can allevi- ate physiological hyperthemia causing damage to the ooeytes during heat stress, heat stress at 41 0C was conducted in the oocytes during in vitro maturation(0 -2 h). The maturation medium (M199) was added with melatonin at a concentration of 1 ×10-9 mol/L, and then the develop- ment of late stages, the number of blastocyst cells, and apoptosis of blastocyst ceils were detected. The results showed that heat stress could sig- nificantly reduce the blastocyst rate and the number of blastoeyst cells of procine oocytes, and the apoptosis rate of blastocyst cells significantly increased. However, the addition of melatonin could alleviate the effect of heat stress on the ooeytes. The results indicate that heat stress can affect oocyte maturation and has an effect on the later development of ooeytes, while the addition of melatonin to the culture medium at a concen- tration of 1 ×10-9 mol/L can alleviate heat -stress damage.
