蝉蜕超微粉抗变态反应作用及初步机制研究

摘　　要：为了探讨蝉蜕超微粉抗变态反应的作用及其初步机制,试验分别进行小鼠被动皮肤过敏反应(PCA)试验、接触性过敏性皮炎(ACD)试验和化合物48/80诱导的小鼠全身过敏反应试验,选择健康Balb/c小鼠,设置空白组、模型对照组、阳性对照组和蝉蜕超微粉高剂量组(100 mg/kg)、中剂量组(50 mg/kg)和低剂量组(10 mg/kg),观察不同剂量蝉蜕超微粉对三种过敏反应的抑制作用,并对PCA、ACD试验中的小鼠过敏部位进行组织切片,光镜下观察肥大细胞的聚集情况,并制备各组含药血清,观察药物对体外培养的P815肥大细胞脱颗粒及组胺释放的影响。结果表明:不同剂量蝉蜕超微粉对小鼠PCA、ACD和化合物48/80诱导的全身过敏反应均有不同程度的抑制作用;不同剂量的蝉蜕制品对体外培养的P815肥大细胞的脱颗粒及组胺释放有一定抑制作用。说明蝉蜕超微粉有明显的抗变态反应作用,且该作用与抑制肥大细胞释放组胺有关。To investigate.the anti - allergic effects of ultrafine powder of Periostracum cicadae （ PC ） and its preliminary mechanism, a series of tests were conducted for mice, including passive cutaneous anaphylaxis （PCA） test, allergic contact dermatitis （ACD） test, and systemic anao phylaxis induced by compound 48/80, respectively. Healthy male Balb/c mice were randomly divided into a blank group, model control group, positive control group, high- dose PC group with 100 mg,/kg ultrafine powder, middle- dose PC group with 50 mg/kg ultrafine powder, and low - dose PC group with 10 mg/kg ultrafine powder. The inhibitory effects of different doses of PC ultrafine powder were observed on the three allergic responses, and the specimens of blue spotted skin from mice in PCA and the pieces of mice ears in ACD were made into tissue sections. The aggregation of mast cells was observed under an electron microscopy, and then drug - contained serum in various groups was prepared, while the effects of the drug were observed on the degranulation and histamine release of P815 ceils cultivated in vitro. The results showed that different doses of PC uhrafine powder had different degrees of inhibitory effects on PCA, ACD, and systemic anaphylaxis induced by compound 48/80 in mice model, and different doses of PC products had a certain inhibitory effect on the degranulation and histamine release of P815 cells cultivated in vitro. The results indicate that the ultrafine powder of Periostracum cicadae has obvious inhibitory effect on allergic reaction, which is related to inhibiting the histamine release of mast cells.
