和田青驴线粒体DNA中CO1基因的PCR扩增与序列分析

摘　　要：为了获得和田青驴线粒体DNA中细胞色素氧化酶(CO1)基因序列,试验采用PCR技术及BLAST在线平台方法对和田青驴血液线粒体DNA中CO1基因进行扩增与序列同源性分析。结果表明:DNA核苷酸序列与AJ009780.1的同源性为99.0%,与KC763190.1、CR388000.2、AB033487.1同源性为98.0%,确认是试验所需要的目的基因。To obtain the sequence of the cytochrome oxidase 1 （CO1） gene in mitochondrial DNA of Hetian Qing donkey, PCR technique and BLAST online platform were used for PCR amplification and sequence homology analysis of the CO1 gene in mitochondrial DNA in blood of He- tian Qing donkey. The results showed that the DNA nucleotide sequence of the CO1 gene shared 99.0% homology with AJ009780.1, and also shared 98.0% homology with KC763190.1, CR388000.2, and AB033487.1. The CO1 gene was confirmed to be the required target gene in the test.
