摘 要:目的探讨胎盘性激素结合球蛋白(sex hormone-binding globulin,SHBG)基因启动子(TAAAA)n重复多态与妊娠期糖尿病(gestational diabetes mellitus,GDM)的关系。方法选择2011年1月至2013年2月在中国医科大学附属盛京医院单胎分娩女婴GDM孕妇129例(GDM组)和健康孕妇119倒(对照组),采用基因片段分析加测序法检测胎盘SHBG基因启动子(TAAAA)n重复多态长度和重复次数,两组间进行比较。结果两组孕妇胎盘均检测到(TAAAA)n重复次数为6、7、8、9、10次重复5个等位基因,6/6、6/7、6/8、6/9、6/10、7/7、7/8、7/9、7/10、8/8、8/9、8/10、9/9、9/10共14种基因型。两组间5种等位基因构成比及各等位基因频率差异均无统计学意义(P〉0.05)。GDM组胎盘长链重复基因型频率虽然高于短链,但差异无统计学意义(x2=2.149,P=0.143)。结论胎盘SHBG基因启动子(TAAAA)n重复多态与妊娠期糖尿病无关。Objective To investigate the relationship between (TAAAA)n repeat polymorphism in the promoter of the sex hormone- binding globulin (SHBG) gene in placental tissue and gestational diabetes mellitus (GDM).Methods GeneScan method was used to detect and identify (TAAAA) n repeat number (alleles) and genotypes for 119 controls and 129 GDM patients who delivered girls in Shengjing Hospital. Results Five alleles containing (TAAAA) 6,7,8,9, 10 repeats and 14 genotypes including 6/6,6/7,6/8,6/9,6/10,7/7,7/8,7/9,7/10,8/8,8/9,8/10,9/9,9/10 repeats geno- types were present in the subjects.There were no significant differences in alleles constituent ratio and frequency be- tween the experimental group and control group.Although the long chain repeat genotype frequency exceeded the short in GDM group, there were no significant differences(x2=2.149,P=0.143). Conclusion (TAAAA)n repeat polymorphism in the promoter of the sex hormone- binding globulin (SHBG) gene in placental tissue has no connection with gestation- al diabetes mellitus.
