肺金生方通过调控MMP-2/TIMP-2平衡抗肺癌侵袭转移研究

摘　　要：目的:探讨肺金生方对多种肺癌细胞株的体外抗肿瘤转移作用。方法:以肺金生方含药血清作用于肺癌A549、PG和PAa细胞,普通倒置显微镜观察各组细胞的生长特点及形态,MTT法检测细胞增殖情况,Boyden小室法观察细胞迁移,ELISA法检测25%肺金生方含药血清对MMP-2、TIMP-2表达的影响。结果:肺金生方含药血清作用于各细胞后,与正常细胞相比形态发生明显改变,明显梭形化,细胞数量减少;各浓度肺金生方组抑制率与空白组相比有差异,其中25%含药血清与空白组相比有显著性差异(P<0.01),25%肺金生方含药血清组能减弱A549、PG和PAa细胞迁移,同时,各细胞肺金生方组MMP-2、TIMP-2的表达与空白血清组相比有差异(P<0.05)。结论:肺金生方可以通过调控MMP-2/TIMP-2平衡达到抗肺癌转移作用。Objective： To study the effect of Feijinsheng Decoction on anti-tumor and anti-metastasis in a variety of lung cancer cell lines in vitro. Methods： Feijinsheng containing serum was used to culture A549 cells in vitro. The morphological changes of cells were observed with phase contrast microscope and cell proliferation was detected by MTT method and the cell migration was observed by Boyden chamber and the expressions of MMP-2 and TIMP-1 were detected by ELISA. Results： Feijinsheng containing serum could cause arrangement disorder in the normal cells,obviously spindle-shaped,the number of cells were reduced. Compared with the control group,the inhibition rate of Feijinsheng containing serum group was different. 25% drug containing serum compared with the blank group had significant difference（ P 〈0. 01） and can abate A549,PG and PAa cells migration. At the same time,the expressions of MMP-2 and TIMP-2compared with the blank serum group＇s were different（ P 0. 05）. Conclusion： Feijinsheng Decoction can regulate the MMP-2 / TIMP-2 balance to anti lung cancer metastasis.
