TLR9和VDR基因多态性与结核病易感性的相关性分析

摘　　要：目的探讨TLR9（rs352139）和VDR基因（rs731236和rs2228570）单核苷酸多态性（SNP）与结核病易感性之间的相关性。方法选取2013年1月-2015年6月期间我院确诊为初治活动性肺结核患者300例（结核组）和300例健康体检者（健康对照组）,采用聚合酶链反应结合高分辨率熔解曲线对rs352139、rs731236和rs2228570位点的SNP进行检测。结果 TLR9（rs352139）的AA和GG基因型比AG基因型人群患结核病的风险增加,OR值分别为1.061（0.76-1.49）和1.472（0.95-2.27）;Taq（rs731236）的TT和CC基因型对比TC基因型人群患结核病的风险增加,OR值分别为1.049（0.68-1.61）和1.204（0.36-3.99）;Fok（rs2228570）的CC和TT基因型对比TC基因型人群患结核病的风险增加,OR值分别为1.164（0.83-1.64）和1.048（0.69-1.61）,但两组间比较差异均无统计学意义（P〉0.05）。结论在中国沈阳地区,TLR9（rs352139）和VDR基因（rs731236和rs2228570）SNP可能与结核病易感性关联不大。Objective To explore the correlation between single nucleotide polymorphism of TLR9（rs352139） and VDR（rs731236 and rs2228570） and susceptibility of tuberculosis. Methods 300 diagnosis as the beginning of active tuberculosis patients and 300 healthy examined people from January 2013 to July 2015 of our hospital were selected.The single nucleotide polymorphism of rs352139,rs731236 and rs2228570 loci were tested by using polymerase chain reaction combined with high resolution melting curve. Results The tuberculosis risk increased in AA and GG genotype of TLR9（rs352139） than AG genotype people,and the OR value were respectively 1.061（0.76-1.49） and 1.472（0.95-2.27）.Thetuberculosis risk increased in TT and CC genotype of Taq（ rs731236） than TCgenotype people,and the OR value were respectively 1.164（0.83-1.64） and 1.048（0.69-1.61）,and the difference had no significant difference（P〉0.05）. Conclusion In Shenyang of China,single nucleotide polymorphism of TLR9（rs352139） and VDR（rs731236 and rs2228570） may have correlation with susceptibility of tuberculosis.
