Study on Therapeutic Effect of Notopterygium Incisum Angelica Pubescens and Pair of Them on Adjuvant Arthritis

摘  要:目的:探讨常用药对羌活与独活及其单味药治疗佐剂关节炎的作用,为临床羌活、独活配伍用药方式提供实验依据。方法:建立完全弗氏佐剂关节炎模型;造模成功后随机分空白组、模型组、阳性组、羌活组、独活组、羌独活组;观察大鼠机械疼痛阈值、足趾容积变化;酶联免疫吸附试验检测足趾组织研磨液IL-1β、IL-6、TNF-α、PGE2水平。结果:给药第14天后,给药组与模型组相较,大鼠对机械压力刺激的痛阈值显著增大(P<0.05),大鼠的足趾容积显著减小(P<0.05),各给药组均能够显著抑制IL-6、TNF-α、PGE2的水平(P<0.05),但是,羌活、独活组对IL-1β水平无影响,羌独活组能够显著减少IL-1β的产生(P<0.05)。结论:药对羌活与独活及其单味药能够在不同程度上显著性的减轻关节炎大鼠的足趾肿胀,提高机械压力刺激的痛阈值,其抑制IL-1β、IL-6、TNF-α、PGE2的异常分泌可能是治疗佐剂关节炎的机制之一。Objective:Explore therapeutic effect of the commonly used drugs of Notopterygium incisum,Angelica pubescens and pair of them on adjuvant arthritis to provide a reliable basis for clinic. Methods:The mode of complete Freund's adjuvant arthritis was established;Six group,control group,model group,positive group,group of Notopterygium incisum,Angelica pubescens and pair of them,were established;mechanical pain thresholds and volume of toe were measured in rats;Levels of IL-1β,IL-6,TNF-α,PGE2 were determined by ELISA. Results:At the 14 th,Mechanical pain threshold was increased in treatment group compared with the model group(P〈0.05),and the volume of the toe was reduced significantly(P〈0.05);The abnormal secretion of IL-6,TNF-α and PGE2 were significantly inhibited in each treatment group(P〈0.05);IL-1β levels are not affected on groups of Notopterygium incisum and Angelica pubescens,however,which was significantly reduced in drug pair of them(P〈0.05). Conclusion:Mechanical pain threshold was increased,the volume of the toe was reduced significantly,IL-1β,IL-6,TNF-α and PGE2 were significantly inhibited,varying degrees in Notopterygium incisum,Angelica pubescens and pair of them,which could be one of the mechanisms of adjuvant arthritis treatment.
