摘 要:目的:通过检测瘦素(Leptin)-神经肽Y(NPY)/血管活性肠肽(VIP)/胃泌素(GAS)信号传导通路及胸腺指数、脾指数的变化,探讨不同浓度黄芪建中汤对脾气虚证大鼠的影响。方法:复制脾气虚证大鼠模型(大黄泻下、游泳力竭和饥饱失常三因素复合法),造模大鼠随机分为正常对照组,模型组,四君子汤组,黄芪建中汤低、中、高剂量组,在用药21 d后检测黄芪建中汤对大鼠血清学及组织学Leptin、NPY、VIP、GAS蛋白表达及胸腺指数、脾指数的影响。结果:与模型组比较,黄芪建中汤中、高剂量组血清木糖浓度、CPK、GAS、NPY含量上升,Leptin及VIP含量下降,与模型组比较差异均有统计学意义(P<0.01或P<0.05),与对照组比较差异有统计学意义(P<0.01或P<0.05),尤以中剂量组作用最为明显。在造模成功后,大鼠组织学Leptin、VIP表达增加,GAS、NPY表达降低,同时伴随指数、胸腺指数的降低(P<0.01)。在给予黄芪建中汤治疗21 d后,黄芪建中汤中、高剂量组Leptin、VIP表达降低,GAS、NPY表达增加,胸腺指数、脾指数也相应回升,模型组比较,差异有显著统计学意义(P<0.01),且中剂量组具有最佳的剂量-效应比(P<0.05或P<0.01)。结论:经方黄芪建中汤治疗脾气虚证的机制可能是通过LeptinNPY/VIP/GAS信号转导通路及提高胸腺指数、脾指数发挥作用,且具有明显的剂量效应关系。Objective:To investigate the effects of Huangqi Jianzhong Decoction with different concentrations on Leptin-NPY/VIP/GAS signal transduction pathway and thymus index,spleen index in spleen-qi deficiency rats. Methods:Spleen-qi deficiency syndrome was induced in rats by compound methods which including Radix et Rhizoma Rhei,exhausting and hunger. Spleen-qi deficiency rats were randomly divided into the normal group,model group,the Four Mild Drugs Decoction group and the low,medium and high dosage of HQJZD group,after 21 d with drug,detection the protein expression of Leptin,NPY,VIP,GAS of Serological and histological and thymus index,spleen index in rats. Results:As compared with that in model control group,the serum xylose concentration,CPK,GAS,NPY contents were rise in the medium and high dosage groups,Leptin and VIP content were decreased,compared with the model group,he differences were statistically significant(P〈0.01 or P〈0.05),there was statistically significant difference compared with the control group(P〈0.01 or P〈0.05),especially in the the middle dose group for the most obvious role. After the success of modeling,histological expression of Leptin and VIP increased,GAS and NPY decreased,accompanied by lower index and thymus index(P〈0.01). After given HQJZD of 21 days,expression of Leptin and VIP decreased,GAS and NPY increased in medium and high dose group,accompanied by higher thymus index and spleen index,compared with model group,the difference was statistically significant(P〈0.01),and medium dose group has the best dose- effect ratio(P〈0.05 or P〈0.01). Conclusion:the intervention mechanism of HQJZD on spleen-qi deficiency syndrome may be play a role through Leptin-NPY/VIP/GAS signal transduction pathway and increase the thymus index and spleen index,but also has an obvious dose-effect relationship.
