摘 要:目的利用原生质体诱变方法从原始菌株炭团菌NX9选育石杉碱甲高生产菌株。方法对菌株NX9进行原生质体紫外诱变,筛选到的正突变菌株进行原生质体NTG进行诱变;结果通过对菌株NX9进行原生质体紫外诱变筛选到正突变株U50,石杉碱甲产量达到19.89μg·L^(-1),比出发菌株高16.7倍。对U50进行原生质体NTG诱变,选育到正突变菌株N65,产量达32.56μg·L^(-1),比出发菌株U50高出0.64倍,比NX9的产量提高了28.07倍,经5次传代后,遗传稳定性良好,可供进一步研究和育种。OBJECTIVE To increase the production of HupA, high HupA-producing strain screened through protoplast mutation from original strain NX9. METHODS UV induced mutagenesis of protoplast was performed on strain NX9,then mutant strains were screened by protoplast NTG mutagenesis. RESULTS Positive mutants strain, U50 was screened by NX9 protoplast UV mutation,which the production of huperzine was reached 19. 89μg · L^-1, 16. 7 times higher than the original strain. CONCLUSION Through the protoplast NTG mutagenesis of U50, positive mutant strain of N65 was obtained, which output can reach 32. 56μg · L^-1 higher than the original strain U50 0. 64 times and 28.07 times more than NX9, after 5 batches, which show that the heredity of strain was well, and the strain can be researched future.
