摘 要:目的探讨三黄片对糖尿病大鼠肾脏细胞间黏附分子1(ICAM1)及血管细胞间黏附分子1(VCAM1)表达的影响。方法制备糖尿病大鼠模型,成模后,用简单随机抽样方法抽取20只分为模型组(10只)和三黄片组(10只),并设立正常组(10只)。三黄片组给予三黄片10 g/kg灌胃,其余两组每日给予同等剂量纯净水灌胃。8周后,取肾脏制备病理切片,链霉菌抗生物素蛋白-过氧化物酶连结法免疫组织化学染色,观察ICAM1及VCAM1的阳性表达。结果正常组大鼠肾质量和肾质量/体质量分别为(1.79±0.20)g和(3.81±0.49)×10^(-3),模型对照组大鼠肾质量和肾质量/体质量分别为(2.29±0.27)g和(8.35±0.68)×10^(-3),三黄片组大鼠肾质量和肾质量/体质量分别为(2.01±0.14)g和(6.08±0.69)×10^(-3)。模型对照组及三黄片组大鼠肾质量和肾质量/体质量显著高于正常组(P<0.05),三黄片组大鼠肾质量和肾质量/体质量显著低于模型对照组(P<0.05)。正常组大鼠尿素氮和肌酐为(7.09±1.03)mmol/L、(42±6)μmol/L,模型对照组大鼠尿素氮和肌酐分别为(10.14±1.13)mmol/L、(56±8)μmol/L,三黄片组大鼠尿素氮和肌酐为(9.33±1.08)mmol/L、(49±7)μmol/L。模型对照组和三黄片组大鼠较正常组大鼠尿素氮和肌酐显著升高(P<0.05)。结论三黄片能明显抑制糖尿病大鼠肾脏ICAM-1及VCAM1表达,具有保护糖尿病大鼠肾脏的作用。Objective To explore influence of Sanhuang tablets on renal expression of intercellular adhesion molecule 1 ( ICAM1 ) and vascular cellular adhesion molecule 1 ( VCAM-1 ) in diabetes mellitus (DM) rats. Methods The DM rat model was made. 20 rats were randomly selected and divided into model group(10 rats) and Sanhuang group( 10 rats) ,and a normal control group( 10 rats) was set up. Sanhuang tablets were given to the Sanhuang group, by 10 g/kg, gavage; the other two groups were given the same amount of pure water,gavage. 8 weeks later,kidneys were removed and the pathologic slices were prepared to observed the expression of ICAM-1 and VCAM-1 by streptavidin-perosidase(SP) immunohistochemical staining. Results Kidney weight and kidney weight/body weight ratio of the normal control group were( 1.79 ±0.20) g,(3.81 ±0.49) ×10^-3,of the model group were (2.29 ±0.27) g,(8.35 ±0.68) ×10^-3,of Sanhuang group were (2.01 ±0. 14) g, (6. 08 ±0.69)×10^-3. Model group and Sanhuang group were significantly higher than the normal control group ( P 〈 0. 05 ), and the Sanbuang group lower than the model group ( P 〈 0. 05 ). Blood urea nitrogen and creatinine of the normal control group were ( 7.09 ±1.03 ) retool/L,(42±6)μmol/L,of the model group were (10. 14 ±1.13) mmol/L, (56 ±8) μmol/L,of the Sanhuang group were (9.33 ±1.08 ) mmol/L, (49 ±7 ) μmo]/L. The model group and Sanhuang group were significantly increased than the normal control group ( P 〈 0. 05 ). Conclusion Sanhuang tablets can inhibit the expression of ICAM-1 and VCAM-1 and protect kidney of the diabetic rats.
