摘　　要：Myogenin是一种诱导肌肉细胞分化和控制肌生成最终分化步骤的重要转录因子。本试验以小鼠成肌细胞(C2C12)为研究材料,采用实时荧光定量PCR(q RT-PCR)、蛋白质印迹(Western Blot)和荧光素酶报告基因分析等方法研究了mi R-186对myogenin基因的调节作用。结果显示,mi R-186在C2C12细胞分化初期表达上调,并能通过作用于myogenin的3'-UTR区显著抑制myogenin的表达。Myogenin is a major transcription factor that induces muscle cell differentiation and controls the final differentiation step of myogenesis.In this study,the murine C2C12 myoblast cell line was used to investigate whether miR-186 is a regulator of myogenin.qRT-PCR, Western Blot and luciferase assays showed that the expression of miR- 186 was increased on the first day after induction of differentiation,and miR-186 repressed myogenin significantly through targeted its 3＂-UTR.
