摘  要:目的:观察电针对脑缺血再灌注损伤大鼠JAK2/STAT3 m RNA及神经元凋亡的影响,探讨电针抗神经元凋亡的机制。方法:将90只雄性SD大鼠随机分为假手术组、模型组、电针组,每组30只,每组按照再灌注时间分为再灌注后2、24、72h 3个亚组,各亚组大鼠均为10只,采取改良大脑中动脉线栓法(MCAO)复制局灶性脑缺血再灌注模型,缺血90min后再灌注。假手术组仅分离暴露右侧颈总动脉不插线栓。模型组和电针组按造模方法造模,造模成功后模型组不进行治疗干预,电针组采用韩氏电针治疗仪,2/15Hz疏密波,刺激百会穴与内关穴,持续30min。各亚组完成相应电针治疗后即断头取脑。运用TUNEL法检测凋亡阳性细胞数,RT-PCR检测JAK m RNA、STAT m RNA表达。结果:再灌注2h后,模型组和电针组之间凋亡阳性粒细胞数、JAK2 m RNA、STAT3m RNA均增高,两组差异无统计学意义,与假手术组比较有显著性差异(P<0.01);再灌注24h、72h后,电针组较模型组差异有统计学意义(P<0.05)。结论:电针能通过抑制脑缺血再灌注损伤后JAK2/STAT3的过度表达达到脑保护、抗神经元凋亡的作用。Objective: To observe the effects of electroacupuncture on JAK2/STAT3 m RNA and neuronal apoptosis of rats with cerebral ischemia/reperfusion injury, and explore the mechanism of anti-apoptosis with electroacupuncture. Methods: 90 male SD rats were randomly divided into sham group, model group and electroacupuncture group, each of which was divided into three subgroups according to reperfusion time(2, 24, 72h), each subgroup for 10 rats. The model of focal cerebral ischemia reperfusion was established by the modii ed middle cerebral artery occlusion(MCAO). The reperfusion was conducted at 90 min after ischemia. In the sham group, the right common carotid artery was merely separated but without being inserted into a thread. The electroacupuncture group was inserted the needles by means of HANS instrument on Baihui acupoint(GV 20) and Neiguan(P6), 2/15 Hz sparse/dense wave, lasting for 30 min. After the treatment, rats in each subgroup were sacrii ced to remove the brain. Then TUNEL apoptosis staining was used to observe neuronal apoptosis and RT-PCR was used to determine the expression of JAK2 m RNA and STAT3 m RNA. Results: At 2h after reperfusion, the number of apoptotic cells and the expression of JAK2 m RNA, STAT3 m RNA were significantly increased in both of the model group and the electroacupuncture group, with there being no significant difference. There were, however, significant differences in the number of apoptotic cells and the expression of JAK2 m RNA, STAT3 m RNA between the sham group and the model group or the electroacupuncture group(P0.01). At 24 h and 72 h after reperfusion, there was significant difference between the model group and the electroacupuncture group(P0.05). Conclusion: The electroacupuncture can protect brain and anti-neuronal apoptosis, which may be related to inhibit the overexpression of JAK2/STAT3 after injury induced by cerebral ischemia and reperfusion.
