摘 要:目的:明确增重剂对茜草饮片质量及其所含大叶茜草素和羟基茜草素含量的影响。方法:采用高效液相色谱法,色谱柱为资生堂Tech Mate CACELL PAK C18柱(250mm×4.6mm,5μm),流动相为乙腈-0.1%三氟乙酸,流速:1.0m L/min,检测波长:240nm,柱温:30℃。结果:大叶茜草素在10-500μg/m L范围内线性良好,羟基茜草素在5-200μg/m L范围内线性良好;两者的平均加样回收率分别为99.32%(RSD=2.26%)和99.59%(RSD=2.63%);实验组茜草的质量均不低于对照组茜草,且对照组茜草中羟基茜草素和大叶茜草素的含量均明显高于实验组。结论:增重剂可增加茜草的质量并明显降低茜草中羟基茜草素和大叶茜草素的含量,同时还通过改变色谱图中各峰的比例影响茜草的质量。Objective: To determine the effects of weighting agents on Radix Rubiae decoction pieces in terms of its weight and contents of mollugin and purpurin. Methods: HPLC was carried on with Shiseido Tech Mate CACELL PAK C18 chromatographic column(250mm×4.6mm, 5μm) and mobile phase of acetonitrile-0.1% tril uoroacetic acid(TFA) under the condition of 1.0m L/min l ow rate, 30℃ of column temperature and 240 nm detection wavelength. Results: The linear ranges of mollugin and purpurin were 10-500μg/m L and 5-200μg/m L, respectively. The average recovery rates of mollugin and purpurin were 99.32%(RSD=2.26%) and 99.59%(RSD=2.63%), respectively. The weight of samples in experimental group was higher than that in control group. In contrast, the content of mollugin and purpurin in experimental group was generally lower than that in control group. Conclusion: Weighting agents increase the weight of samples, but weighting agents could significantly decrease the content of purpurin and mollugin in Radix Rubiae decoction pieces. Meanwhile, the ratio of peaks in HPLC chromatogram is inl uenced by weighting agents that could impact on the quality of Radix Rubiae decoction pieces.
