摘  要:以紫玉兰花粉为材料,采用离体萌发法研究不同培养基组分对紫玉兰花粉萌发和花粉管生长的影响,确定花粉萌发的最佳培养条件,并用25/10℃、15/10℃、有水和无水4种条件交互处理观察紫玉兰单花的开花进程。研究表明:(1)紫玉兰花粉萌发的最佳培养基是3%蔗糖+0.003%~0.005%硼酸+0.02%氯化钙;(2)花粉萌发率和花粉管长度达到最大值为6 h;(3)15/10℃处理的紫玉兰花瓣明显比25/10℃的大;(4)有水处理的紫玉兰花瓣比无水处理的花瓣干重大;(5)有水处理的表皮细胞排列整齐且比无水处理的表皮细胞大;(6)紫玉兰花药开裂散粉需在干燥环境下进行。本研究对延长花期、增大花瓣面积、提高其观赏价值以及杂交育种具有重要意义。The effects of medium components on pollen germination and pollen tube growth of Magnolia lili- flora Desr. were studied using in vitro culture. M. liliflora was subjected to four environmental conditions/treatments (25/10℃, 15/10℃, with water or without water) for flowering. The results showed that the best medium for pollen germination was 3% sucrose, 0.003% -0.005% boric acid plus 0.02% calcium chloride. The optimum time was 6 h for pollen germination and pollen tube growth. The petals were obviously bigger in the 15/10℃ treatment than that in the 25/10~C treatment. The dry weight of petals was higher in the water treatment than that of the water-free treatment. The epidermal cells appeared to be bigger and much uniform in the water treatment. Anther dehiscenco required a dry environment. This research shows significance for improving the ornamental value and facilitating cross breedin~ ofM. lilillora with extended blooming period and enlarged oetals.
