摘 要:为了得到产气荚膜梭菌β毒素单克隆抗体,以重组产气荚膜梭菌β毒素为抗原免疫小鼠,取其脾细胞与骨髓瘤细胞SP2/0融合并筛选出阳性单克隆杂交瘤细胞株,接种至小鼠腹腔,培养后抽取腹水并用正辛酸-硫酸铵纯化法进行纯化,从而获得抗产气荚膜梭菌β毒素单克隆抗体。该单克隆抗体经特异性试验,能与天然β毒素及重组β毒素发生特异性反应,且效价较高,为进一步建立快速有效的临床检测技术奠定了基础。In order to get the monoclonal antibodies of Clostridiumperfringensβ-toxin,the recombinant Clostridium perfringensβ-toxin was used as antigen to immunize mice.After cell fusion between spleen cells and SP2/0,the positive hybridoma cells were screened.The positive monoclonal hybridoma cell lines were inoculated intraperitoneally into mice.The monoclonal antibodies againstβ-toxin of Clostridium perfringens were obtained,which were purified by octanoic-ammonium sulfate purification method.The specificity experiments showed that the monoclonal antibodies specifically reacted with recombinantβ-toxin and crude naturalβ-toxins,and the potency was high.The study laid the foundation for establishing a rapid and effective method for clinical test.
