鹿红颗粒对心力衰竭大鼠ACE2-Ang(1-7)轴的影响

摘　　要：目的:评价鹿红颗粒对心肌梗死后心力衰竭(CHF)模型大鼠血浆ACE-AngⅡ轴及ACE2-Ang(1-7)轴的影响。方法:40只SD大鼠随机分为模型组、假手术组、雅施达组及鹿红颗粒组,以结扎左冠状动脉的方法制备心肌梗死后心力衰竭模型。药物干预8周后记录各组大鼠心脏重量,计算心脏重量与体质量的比值(HW/BW)以及左心室重量指数(LVMI);HE染色观察各组大鼠心肌病理改变;酶免疫分析法检测血浆ACE、ACE2、AngⅡ及Ang(1-7)水平。结果:与模型组相比,鹿红颗粒组HW/BW及LVMI明显降低(P<0.01),心肌病理改变显著减轻,血浆ACE水平未见明显下降(P>0.05),但ACE2和Ang(1-7)水平明显升高(P<0.01),AngⅡ水平显著降低(P<0.01);与雅施达组比较,鹿红颗粒组ACE2和Ang(1-7)水平明显升高(P<0.01),AngⅡ水平未见明显差异(P>0.05),Ang(1-7)/AngⅡ显著升高(P<0.01)。结论:鹿红颗粒能显著提高CHF大鼠ACE2-Ang(1-7)轴的水平、降低AngⅡ水平,从而抑制心室重构、减轻心肌损伤。Objective： To observe the effect of＂Luhong Granule＂on ACE-Ang Ⅱ and ACE2-Ang（ 1-7） axis in rats with heart failure. Methods： 40 heart failure rats were randomly allocated into model group,sham operation group,perindopril group and＂Luhong Granule＂group,and they were established by ligating left anterial descending coronary artery. 8 weeks after medicine administration,the rats from each group were detected for heart weight. HW / BW and LVMI were calculated; HE staining was used for histopathology test. We used enzyme linked immunosorbent assay to detect plasma ACE,ACE2,Ang Ⅱand Ang（ 1-7） levels. Results： Compared with model group,＂Luhong Granule＂can obviously lower HW / BW and LVMI（ P 〈0. 01）; ＂Luhong Granule＂can obviously alleviate myocardium histopathology impact. Plasma ACE level was not changed（ P 〉0. 05）,while ACE2 and Ang（ 1-7） levels were significantly improved and Ang Ⅱ level decreased（ P〈 0. 01）.Compared with perindopril group,plasma ACE2 and Ang（ 1-7） levels were significantly improved in＂Luhong Granule＂group（ P 〈0. 01）. The Ang Ⅱ level was roughly the same in two groups（ P 〉0. 05）. The value of Ang（ 1-7） /Ang Ⅱ was significantly higher in＂Luhong Granule＂group than that in perindopril group（ P 〈0. 01）. Conclusion： ＂Luhong Granule＂can improve the plasma ACE2-Ang（ 1-7） axis and lower Ang Ⅱ level in rats with heart failure,and thereby to inhibit ventricular remodeling and alleviate myocardium impact.
