双黄升白颗粒对化疗所致骨髓抑制Lewis肺癌荷瘤鼠Notch信号通路的双重调控作用  被引量:6

摘  要:目的:探讨双黄升白颗粒对荷瘤鼠化疗骨髓抑制期Notch信号通路的调控作用。方法:采用Lewis肺癌荷瘤鼠腹腔注射环磷酰胺300 mg/kg制成化疗骨髓抑制模型。荷瘤鼠随机分为空白组、模型组、治疗组,另设模拟接种的正常组,每组8只。治疗组在造模同时灌服双黄升白颗粒溶液0.4 ml/d,连续6 d;其他组均灌服生理盐水0.4 ml/d,连续6 d。常规计数白细胞、红细胞、血小板数和骨髓有核细胞数,称取瘤重,荧光定量PCR法检测Notch信号通路中Notch1、Notch2、Notch3、Jagged、CSL的mRNA表达。结果:治疗组白细胞数、骨髓有核细胞数均高于模型组(P<0.05),Notch1、Notch2、Notch3、CSL在骨髓中的mRNA表达高于模型组(P<0.05),在肿瘤中的表达则低于模型组(P<0.05);各组红细胞、血小板、瘤重、Jagged mRNA表达未发现明显差异(P>0.05)。结论:双黄升白颗粒治疗骨髓抑制的作用机制可能与调控Notch信号通路有关,且该调控作用具有双重特性,即在上调骨髓Notch信号通路主要基因表达的同时抑制肿瘤中部分基因表达。Objective: To discuss effect of "Shuanghuang Shengbai Granules"( SHSB) on tumor-bearing mice with chemotherapy induced myelosuppression by regulating Notch signaling pathway. Methods: Chemotherapy induced myelosuppression model of lewis lung tumor-bearing mice by intraperitoneal injection of cyclophosphamide 300 mg / kg was set up. The mice were randomized into 4 groups: blank group,model group,treatment group and normal control group,8 in each group. The treatment group was perfused SHSB with 0. 4 ml,once a day,for successive 6 days. General counting white blood cells,red blood cells,platelet,bone marrow cell count and tumor size,fluorescence quantitative PCR method was used to detect major genes mRNA expression of Notch signaling pathway( Notch1,Notch2,Notch3 m,Jagged,CSL). Results: The number of white blood cells and bone marrow cell in treatment group was higher than that in the model group( P 〈0. 05),Notch1,Notch2,Notch3 and CSL mRNA expression in the bone marrow of treatment group was higher than that in the model group( P 〈0. 05),lower in tumor than that in the model group( P 〈0. 05). There was no significant difference of erythrocytes,platelets expression,tumor size and Jagged mRNA expression in each group( P〉 0. 05). Conclusion: The mechanism of SHSB granules' effect on myelosuppression was related to regulating Notch signaling,and it has dual regulation effect on Notch signaling pathway by increasing expression of bone-related genes while inhibiting the expression of some genes in tumors.
