丹蛭降糖胶囊对糖尿病大鼠胰腺NLRP3炎症小体的影响

摘　　要：目的:观察具有益气养阴活血作用的丹蛭降糖胶囊(DJC)对糖尿病大鼠胰腺炎症小体NLRP3的影响。方法:取大鼠55只,除正常对照组9只外,其余大鼠高能量饮食4周后加小剂量一次性腹腔注射STZ的方法建立2型糖尿病大鼠模型;造模成功后,模型组大鼠分为模型组,DJC高、低剂量组和吡格列酮组。各药物组分别予DJC(1.08、.0.54 g·kg-1·d-1)、吡格列酮10 mg·kg-1·d-1,正常组、模型组予等量生理盐水灌胃干预,连续8周。检测大鼠血糖及血脂水平;实时荧光定量PCR方法检测大鼠胰腺组织NLRP3mRNA表达。结果:与正常对照组比较,模型组胰腺炎症小体NLRP3 mRNA表达明显升高(P<0.01)。与模型组比较,DJC高剂量组NLRP3 mRNA表达明显降低(P<0.01);DJC高、低剂量组及吡格列酮组的血糖和血脂水平显著降低(P<0.01),而各用药组之间比较无明显差异。结论:DJC可控制NLRP3的过度表达,抑制细胞炎症反应、改善微炎症状态,从而减轻氧化应激损伤、保护胰岛B细胞。Objective： To investigate the effects of＂Danzhi Jiangtang Capsule＂on NLRP3 inflammasome of pancreas in diabetic rats. Methods： 55 rats except the 9 rats in control group in the experimental group were used to set models by method of feeding with high calorie diet for 4 weeks and then injected with streptozocin（ STZ）. When diabetes rats model was successfully built,the animals in the experimental group were divided into model group,high and low dose＂Danzhi Jiangtang Capsule＂groups, and pioglitazone group. High and low dose ＂Danzhi Jiangtang Capsule ＂ groups were treated with1. 08 g /（ kg·d） and 0. 54 g /（ kg·d） by gavage,pioglitazone group was treated with 10 mg /（ kg·d） by gavage,the control group and the model group were treated with equal quantity physiological saline. After 8 weeks,the levels of blood glucose and lipid were detected,and the mRNA expressions of NLRP3 inflammasome were detected by the method of PCR. Results：Compared with the normal control group,the mRNA expressions of NLRP3 inflammasome increased significantly（ P〈 0. 01）;compared with model group,the mRNA expressions of NLRP3 inflammasome decreased obviously in high dose ＂Danzhi Jiangtang Capsule＂group（ P〈 0. 01）. Compared with model group,the levels of blood glucose and lipid decreased obviously in high and low dose ＂Danzhi Jiangtang Capsule ＂ groups and pioglitazone group（ P 〈0. 01）,and there is no statistical significance in each group. Conclusion： ＂Danzhi Jiangtang Capsule＂can control the overexpression of NLRP3 inflammasome,restrain cells inflammatory response and improve microinflammatory state thereby to alleviate oxidative stress injury and protect pancreas B cells.
