摘 要:目的 :观察桂枝汤对糖尿病模型大鼠心脏自主神经损伤的治疗作用。方法 :55只雄性Wistar大鼠,其中10只为空白对照组,45只腹腔注射链脲佐菌素(STZ 60 mg/kg)诱导糖尿病模型,成模4周后随机分为模型对照组、弥可保组、桂枝汤组,灌胃治疗4周。Elisa法检测室间隔匀浆中酪氨酸羟化酶(TH)、乙酰胆碱酶转移酶(Ch AT)、神经生长因子(NGF)、生长相关蛋白(GAP-43)、睫状神经生长因子(CNTF)及肿瘤坏死因子(TNF-α)的含量。结果:模型对照组TH、NGF、GAP-43、TNF-α明显升高(P<0.01),Ch AT、CNTF明显下降(P<0.01),TH/Ch AT比值升高(P<0.01);与模型对照组比较,桂枝汤组Ch AT、GAP-43、CNTF升高(P<0.05),TH、NGF、TNF-α下降(P<0.01),TH/Ch AT比值明显下降(P<0.01)。结论 :糖尿病造成心脏自主神经损伤,交感神经亢进,迷走神经受损。桂枝汤能促进迷走神经的修复,降低亢进的交感神经张力,维持交感-迷走神经的平衡。Objective:To observe the curative effect of Guizhi decoction on cardiac autonomic nerves damage of model rats with diabete. Methods:Fifty- five male Wistar rats were selected as the experimental objects.10 of them were assigned to the normal group and the other 45 were given intraperitoneal injection of streptozocin(STZ,60 mg/kg) to induce diabetes mellitus(DM) model. After 4 weeks of modeling,the rats were radomly divided into the model control group,methycobal group and Guizhi decoction group. The medicine were intragastrically administrated for 4 weeks. Ventricular septal homogenate was taken to detect the levels of tyrosine hydroxylase(TH),choline acetyltransferase(Ch AT),nerve growth factor(NGF),growth- associated protein(GAP- 43),ciliary neurotrophic factor(CNTF) and tumor necrosis factor(TNF- α) by Elisa.Results:Compared with the normal rats,the DM rats had higher levels of TH,NGF,GAP- 43 and TNF- α(P0.01) and lower levels of Ch AT and CNTF(P 〈0.01). The ratio of TH/Ch AT of DM rats increased(P 〈0.01).Compared with the model control group,the levels of Ch AT,GAP- 43 and CNTF of the Guizhi decoction group increased(P 〈0.05) and the levels of TH,NGF and TNF- α decreased(P 〈0.01). The ratio of TH/Ch AT of the Guizhi decoction group significantly decreased(P 〈0.01). Conclusion:Dabetes can lead to cardiac au tonomic nerves damage,sympathetic nerve hyperactivity and vagus damage. Guizhi decoction can promote the recovery of the vagus,decrease the hyperactive sympathetic tone and keep the balance between sympathetic and vagus nerve.
