摘  要:目的探讨薯蓣皂苷对胶原诱导型关节炎小鼠的治疗作用及其机制。方法选用DBA1/J小鼠建立鸡Ⅱ型胶原诱导型关节炎小鼠模型(CIA),将造模成功的小鼠采用随机数字表法分为CIA模型组、薯蓣皂苷组、雷公藤多苷组,另设正常对照组,药物组于初次免疫后第21天开始灌胃给药,对照组和模型组给相同体积溶媒。初次免疫第35天处死小鼠,无菌取腹股沟淋巴结制成单细胞悬液,用实时荧光定量PCR技术检测Th17、Treg细胞特异性转录因子RORγt、Foxp3的表达情况。结果模型组与对照组相比,RORγt显著升高(P<0.05);2药物组与模型组相比RORγt显著下降(P<0.05);与模型组相比,薯蓣皂苷组Foxp3显著升高(P<0.05)。结论薯蓣皂苷可以调节Th17、Treg细胞特异转录因子RORγt、Foxp3的平衡。This study performed to observe the effects of Dioscin on the chicken type Ⅱ collagen-induced arthritis(CIA) and investigate the underlying mechanism of Dioscin in treating rheumatoid arthritis(RA). DBA1/J mice were chosen as experimental objects, and randomly divided into 4 groups: model, dioscin, triptolide group, and normal control group. For drug groups(dioscin group and triptolide group), drugs were administered 21 d after the first immunization; for the normal control group and the model group, saline was administered. 35 d after the first immunization, inguinal lymph nodes were collected sterilely and made into single cell suspension for detecting the expressions of Th17 and Treg cell specific transcription factor RORγt and Foxp3 using real-time PCR technique.Data revealed that RORγt in model group was increased significantly(P〈0.05) compared with the normal control group, RORγt decreased significantly in the drug groups(P〈0.05) compared with the model group, and Foxp3 increased significantly in the dioscin group(P〈0.05) compared with the model group. In conclusion, the dioscin can balance the Th17 and Treg cell specific transcription factor RORγt and Foxp3.
