摘 要:为探讨外源小剂量IFN-γ对流产大鼠的免疫调控作用,分别采用反转录-聚合酶链式反应(reverse transcription-polymerase chain reaction,RT-PCR)法、免疫组化法和酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)法研究了流产大鼠子宫和卵巢IL-18 mRNA蛋白的动态表达和细胞定位,并检测了IL-18在外周血中的动态变化。结果表明:IL-18 mRNA在正常妊娠组大鼠子宫和卵巢表达最高;在2.4 IU和2.8 IU IFN-γ组表达低于正常妊娠组(P<0.01);2.0 IU组IL-18 mRNA的表达最接近正常妊娠组(P>0.05)。正常妊娠组大鼠卵巢及子宫IL-18蛋白呈强阳性表达;流产组大鼠卵巢和子宫中IL-18蛋白表达低于正常妊娠组(P<0.01);2.0 IU IFN-γ组IL-18蛋白表达接近正常妊娠组(P>0.05)。流产组大鼠外周血中IL-18表达低于正常妊娠组(P<0.01),也低于2.0 IU、2.4 IU和2.8 IU IFN-γ组(P<0.05)。说明外源性小剂量的IFN-γ可通过调控妊娠早期大鼠子宫、卵巢及外周血IL-18的水平调节妊娠。外源注射2.0 IU IFN-γ对于防止胚胎丢失、维持妊娠或预防早期流产有一定的参考价值。To explore the immunological regulatory effect of exogenous low dose IFN-γ on abortion rats,the dynamic expression of IL-18 mRNA and the cellular localization in uterus and ovaries were researched by reverse transcription-polymerase chain reaction( RT-PCR), immunohistochemistry and enzyme-linked immunosorbent assay( ELISA). Then IL-18 dynamic change in peripheral blood of abortion rats was also determined. The results show that the expression of IL-18 mRNA in uterus and ovary of rats in normal pregnancy group is the highest. The expression of IL-18 mRNA in 2. 4 IU and 2. 8 IU IFN-γ groups is lower than that in normal pregnancy group( P < 0. 01). The expression of IL-18 mRNA in 2. 0 IU group is the most close to normal pregnancy group( P > 0. 05). The expression of IL-18 protein in the ovary and uterus in the normal pregnancy group is strongly positive,and the expression of IL-18 protein in the ovary and uterus of the abortion groups is lower than that in the normal pregnancy group( P < 0. 01). The expression of IL-18 protein in the 2. 0 IU IFN-γ group is close to the normal pregnancy group( P > 0. 05). The expression of IL-18 in peripheral blood of abortion group is lower than that in normal pregnancy group( P < 0. 01),and it is also lower than that of 2. 0 IU,2. 4 IU and 2. 8 IU IFN-γ( P < 0. 05). It suggests that the low exogenous dose IFN-γ can control pregnancy by regulating the level of IL-18 in uterus,ovary and peripheral blood of rats during early pregnancy. Effects of exogenous injection 2. 0 IU IFN-γ on the prevention of fetal loss,maintenance of pregnancy and prevention of early pregnancy abortion have a certain reference value.
