摘 要:目的:观察黄芪多糖对环磷酰胺导致大鼠气虚血瘀证的影响。方法:将SD大鼠随机分为6组,黄芪多糖高、中、低剂量组、丹参组、模型组、空白组。以环磷酰胺腹腔注射和生理盐水饲养制作大鼠气虚血瘀证模型。造模后给药组分别腹腔注射给药;模型组腹腔注射生理盐水,连续5天;空白组不作处理。第9天采血检测血液流变学指标、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)及血清内皮缩血管肽(ET-1)、一氧化氮(NO)的含量。结果:模型组在各切变率下的全血粘度、血沉(ESR)、血浆粘度、红细胞压积(HCT)、PT、APTT及ET-1明显高于空白组(P<0.05),说明造模成功。除黄芪多糖低剂量组HCT与模型组比较无统计学意义外(P>0.05),给药组在各切变率下的全血粘度、ESR、血浆粘度、HCT、PT、APTT、ET-1均比模型组明显降低(P<0.05);各给药组NO含量明显高于模型组(P<0.05);黄芪多糖高剂量组在各切变率下的全血粘度明显低于其他各给药组(P<0.05),黄芪多糖高剂量组在降低ET-1水平及升高NO含量方面明显优于其它给药组(P<0.05);黄芪多糖高剂量组APTT较丹参组缩短(P<0.05)。结论:黄芪多糖可确切改善环磷酰胺导致大鼠的气虚血瘀证,以高剂量作用最为明显。其作用机制可能为降低血清ET-1水平及增加NO含量。Objective: To observe the effect of astragalus polysaccharides ( APS ) on Qi - deficiency and blood - stasis in rats induced by cyclophosphamide. Methods : All 96 Sprague - Dawley rats were divided randomly into six groups, namely, APS high, middle,low dosage group, danshen group and blank control, model control group averagely and made them into Qi - deficiency and blood - stasis model with cyclophosphamide intraperitoneal perfusion through 3 days and fed on 0.9% NS. The dosage groups respectively accepted intraperitoneal injection of drug, model control group accepted saline, all for 5 days, blank control group did nothing. Blood were collected on the ninth day to observe the changes on hemorheology and the content of NO and ET - 1. Results:The model group under the shear rate of whole blood viscosity,ESR, HCT, plasma viscosity, PT, APTT were significantly higher than the blank control group ( P 〈 0.05 ), the model was built successfully. Except the APS low dosage group HCT was no statistical difference than the model group(P 〉 0.05 ) , all the dosage groups under the shear rate of whole blood viscosity, ESR, HCT, plasma viscosity, PT, APTT were significantly lower than the model group( P 〈 0.05 ). The APS high dosage group under the shear rate of whole blood viscosity were lower than the other dosage groups, also it increased the content of NO and reduced the level of ET - 1 were obviously better than the other dosage groups, and APTT were lower than danshen group. Conclusion:APS can obviously improve the Qi - deficiency and blood - stasis in rats induced by cyclophosphamide,the possible mechanism may be reducing the levels of ET - 1 and increasing the NO content.
