摘 要:目的:观察柴朴汤含药血清对支气管上皮细胞(bronchial epithelial cells,HBECs)中核转录因子-κB(nuclear transcription factor-kappa B,NF-κB)及Toll样受体-4(Toll-like receptor 4,TLR-4)表达的影响,探讨柴朴汤治疗哮喘的机制。方法:制备柴朴汤含药血清及哮喘血清。将体外培养的气道上皮细胞分为6组进行处理:正常血清组(A组),柴朴汤高剂量血清+哮喘血清组(B组),柴朴汤中剂量血清+哮喘血清组(C组),柴朴汤低剂量血清+哮喘血清组(D组),生理盐水+哮喘血清组(E组),哮喘血清组(F组),采用免疫细胞化学及real-time PCR检测哮喘血清及不同剂量的柴朴汤含药血清对支气管上皮细胞中TLR-4、NF-κB蛋白及mRNA表达的影响。结果:TLR-4、NF-κB蛋白及mRNA在A组表达最弱,经哮喘血清干预后E、F组TLR-4、NF-κB蛋白及mRNA表达均明显增强,与A组比较差异有统计学意义(P<0.05);柴朴汤干预后B、C、D组NF-κB蛋白及mRNA表达下降,与E、F组比较差异有统计学意义(P<0.05)。结论:哮喘血清可诱导气道上皮细胞TLR-4及NF-κB表达明显增强,而柴朴汤含药血清能下调哮喘血清诱导的支气管上皮细胞中TLR-4及NF-κB的表达。Objective: To observed the Chaipu decoction serum's affection for the expressions of Toll-like receptors 4( TLR-4) and nuclear transcription factor-kappa B( NF-κB) in the airway epithelial cells,to investigete the mechanism of Chaipu decoction treatment of asthma. Methods: The Chaipu decoction and asthma serum were preparerd. The in vitro cultured HBECs were randomly divided into six groups: normal serum group( Group A),Chaipu decoction high dose + asthma serum group( Group B),medium Chaipu dose + asthma serum group( Group C),medium Chaipu dose + asthma serum group( Group D),saline serum + asthma serum group( Group E),asthma serum group( Group F). The expressions of the mRNA and protein of NF-κB and TLR-4 in the different groups were respectively detected by RT-PCR and immunohistochemical method. Results: The expression levels of TLR-4,NF-κB mRNA and protein in Group A were in the minimum amount,with asthmatic serum intervention the expression levels of TLR-4,NF-κB mRNA and protein in Group E,F were significantly higher than those in Group A( P〈0. 05). The expression levels of TLR-4,NF-κB mRNA and protein in Group B,C,D( with Chaipu decoction serum intervention) were significantly lower than those in Group E,F( P〈0. 05). Conclusion:Asthmatic secrum can increase the expression of NF-κB and TLR-4 in airway epithelial cells. Chaipu decoction serum can regulate the expressions of TLR-4,NF-κB in airway epithelial cells.
