摘 要:目的借助脂质体仿生膜手段探寻传统中药板蓝根药材里的有效成分。方法采用薄膜分散法制备空白脂质体,将其与板蓝根提取液置于37℃水浴中孵化;运用离心沉淀-超滤法,分离脂质体、样品超滤液。借助HPLC-UV、HPLC-MS以及氨基酸衍生化手段跟踪检测样品被吸收情况,综合分析推断脂质体仿生膜筛选下的板蓝根有效成分。结果通过样品脂质体混悬液、板蓝根提取液、对照品溶液与阴性样品溶液之间的色谱、质谱数据对比分析,确定在板蓝根脂质体中存在5种主要板蓝根成分,分别是精氨酸、胞苷、尿苷、表告依春和腺苷。结论这5种成分可能是中药板蓝根药理作用的主要有效成分,而采用的脂质体仿生膜可以作为一种可行的中药复杂样品筛选手段。Objective To explore effective compounds of Radix isatidis, a traditional Chinese medicine, by liposomebased biomimetic membrane method. Methods The blank liposome which was prepared by film dispersion method, and Radix isatidis extraction solution were mixed under 37 ℃ water bath for the incubation. Liposome and sample ultrafiltrate were separated through centrifugation-ultrafiltration method. The absorption of sample was recorded under HPLC-UV, HPLC-MS and amino acid derivatization, the effective compounds of Radix isatidis were comprehensively analysed and inferred through liposome-based biomimetic membrane method. Results After comparative analysis of chromatography and mass spectrometry data between sample-liposome suspension, Radix isatidis extract solution, reference solution and negative control solution, five major compounds in Radix isatidis was determined as arginine, cytidine, uridine, epigoitrin and adenosine. Conclusion These 5 compounds may be the main effective compounds in Radix isatidis, and this liposome-based biomimetic membrane method can be a feasible screening approach in complex traditional Chinese medicine.
