摘 要:目的筛选王不留行抗炎镇痛的活性部位并研究其抗炎作用机制。方法采用溶剂萃取法将生、炒王不留行的80%乙醇提取物分离为不同部位,筛选其抗炎镇痛的活性部位,检测活性部位对丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)和NO的影响。结果生、炒王不留行的水、正丁醇和乙酸乙酯部位均能极显著减轻二甲苯所致的小鼠耳廓肿胀;炒王不留行的3个萃取部位均能剂量依赖性地抑制小鼠棉球肉芽肿,且乙酸乙酯部位的抗炎效果最优;炒王不留行的3个萃取部位均能剂量依赖性地降低乙酸致小鼠扭体次数和热致小鼠舔足次数,且正丁醇部位的镇痛作用最优。炒王不留行的3个萃取部位均能剂量依赖性地降低慢性炎症小鼠血清中MDA和TNF-α的产生及肝脏组织中NO、总一氧化氮合酶(TNOS)和诱导型一氧化氮合酶(i NOS)的含量。结论炒王不留行抗急性炎症的活性大于生王不留行的;炒王不留行的乙酸乙酯部位的抗炎活性最强,正丁醇部位的镇痛活性最强,其抗炎作用可能与抑制MDA、TNF-α和NO的产生有关。OBJECTIVE To screen the anti - inflammatory and analgesic active fractions from Cowherb seeds and to study the mech- anism of the anti - inflammatory. METHODS Cowherb seeds and fried Cowherb seeds were respectively extracted with 80% EtOH under reflux conditions. The anti - inflammatory and analgesic effective fractions were studied in vivo. MDA, TNF - α and NO contents were detected. RESULTS Except the aether extracts, the acetoacetate, n - butanol and water extracts from the Cowherb seeds andfried Cowherb seeds significantly reduced the xylene - induced ear swelling in mice. The three extracts from the fried Cowherb seeds sig- nificantly reduced the cotton - induced granuloma in a dose - dependent manner, and the anti - inflammatory effect of the acetoaeetate extracts was much better than the other two extracts. The three extracts from the fried Cowherb seeds significantly reduced the acid - induced writhing numbers and heat - induced licking numbers in mice in a dose - dependent manner, and the analgesic effect of n - butanol extracts was much better than the other two extracts. The three extracts from the fried Cowherb seeds significantly decreased the MDA and TNF -α levels in serum of cotton - induced granuloma mice in dose - dependent manner. Furthermore, they inhibited NO, TNOS and iNOS production in liver tissue of cotton - induced granuloma mice in a dose - dependent manner. CONCLUSION The fried Cowherb seeds showed more potent anti - inflammatory effects than the Cowherb seeds in acute inflammation model in mice. The acetoacetate extracts were the strongest anti - inflammatory effective fraction of the fried Cowherb seeds, while the n - butanol extracts were the strongest analgesic effective fraction. The anti - inflammatory mechanism of the Cowherb seeds extracts may be relative to the inhibition of MDA and TNF - α and NO production.
