摘  要:目的:探讨碱性成纤维细胞生长因子(bFGF)和骨髓间充质干细胞(BMSCs)联合移植对脊髓半横断损伤后神经再生的影响。方法:利用血清培养技术获得SD大鼠BMSCs行CM-DiI标记。80只大鼠随机分为4组:正常组、损伤组、BMSCs移植组、bFGF+BMSCs移植组,每组各20只。建立脊髓半横断损伤模型,移植组采用明胶海绵和细胞悬液填充。分别于术后3、7、14、21 d用BBB评分法观察大鼠后肢运动功能的恢复情况,免疫荧光组织化学染色法检测损伤区CM-DiI、GFAP和GAP-43的表达变化,并对其进行相关性分析。结果:bFGF+BMSCs移植组和BMSCs移植组大鼠的左后肢运动功能较损伤组有明显改善(P<0.05),且bFGF+BMSCs移植组与BMSC移植组相比亦有显著性差异(P<0.05);损伤组GAP-43的表达量极少,BMSCs移植组则有少量表达,而bFGF+BMSCs移植组呈高表达;bFGF+BMSCs移植组术后早期GFAP高表达,BMSCs移植组次之,损伤组最少,但14d后,GFAP表达刚好相反。结论:bFGF+BMSCs移植较BMSCs移植能更好地促进脊髓损伤后神经的再生。Objective:To study the effect of basic fibroblast growth factor(bFGF) and bone mesenchymal stem cells(BMSCs) unite transplantation on neural regeneration after spinal cord left-hemisection.Methods:BMSCs were obtained from SD rats by using the serum culturing.BMSCs labeled by chloromethyl-benzamidodialkylcarbocyanine(CM-DiI) and bFGF were transplanted.Eighty rats were divided randomly in to four groups including normal control group,injured group,BMSCs transplantation group and bFGF + BMSCs transplantation group(n = 20 for each group).The rat model of left-hemisection spinal cord was prepared.For transplantation group,the spinal cord defect was implanted with gelfoam and cell suspension.BBB(Basso,Beattie,Bressnahan locomotor rateing scale) were assessed to detect the recoveryof hindlimb functionof rats by blinder observers at 3,7,14 21 d after surgery.Immunofluorescence histochemistry was used to analyze the expression change of CM-DiI,GFAP and GAP-43.Results:Left hind limb function for the two transplantation were showed significantly improved,compared with that of injured group(P 〈0.05),and the bFGF + BMSCs transplantation group has a more significant improvement than the BMSCs transplantation group(P 〈0.05).Immunofluorescencehistochemistry showed that the level of GAP-43 expression was markedly high in the bFGF + BMSCs transplantation group,a certain degree of GAP-43 expression was shows in the BMSCs transplantation group,the injured group showed minimal.Early postoperative,the level of GFAP expression was highest in the bFGF + BMSCs transplantation group,and the level of GFAP expression decreased in the BMSCs transplantation group,the injured group showed lower.However,in 14 d and 21 d the level of GFAP expression was contrary in the three groups.Conclusion:The combination transplantation of bFGF and BMSCs is effective in improving the neural regeneration after spinal cord injury than transplanted with BMSCs alone.
