氯吡格雷对血小板聚集及肝细胞色素P4503A4表达的影响

摘　　要：目的探讨氯吡格雷对大鼠血小板聚集及肝细胞色素P4503A4表达的影响。方法 45只Wistar大鼠按照体重随机分为3组(空白组,对照组和实验组),每组15只。空白组大鼠灌胃生理盐水5 m L·kg-1·d-1,对照组灌胃阿司匹林100 mg·kg-1·d-1,实验组大鼠灌胃氯吡格雷20 mg·kg-1·d-1,均5m L,3组大鼠均处理7 d。药物处理7 d后,心脏取血,检测血小板聚集率和血小板聚集抑制率。Western blot法检测3组肝细胞色素P4503A4表达水平。结果实验组大鼠血小板最大聚集率为(37.23±5.01)%,显著低于空白组的(57.21±5.23)%和对照组的(47.23±4.21)%(P<0.05);实验组大鼠血小板聚集抑制率为(33.56±4.64)%,显著高于对照组的(21.12±3.36)%(P<0.05);与空白组和对照组相比,实验组大鼠肝细胞色素P4503A4表达水平显著降低(P<0.05)。结论氯吡格雷可能通过抑制大鼠肝细胞色素P4503A4表达来达到抗血小板功能的作用。Objective To evaluate the clopidogrel on platelet aggregation and cytochrome P4503A4 expression in rats in order to the mechanism for clopidogrel anti- platelet. Methods Forty- five Wistar rats were randomly divided in to blank group（ n = 15）,control group（ n = 15） and experiment group（ n = 15）,with normal saline 5 m L·kg- 1·d- 1intragastric administration,aspirin 100 mg·kg- 1·d- 1intragastric administration and clopidogrel 20 mg·kg- 1·d- 1intragastric administration with 7days,respectively（ all 5 m L）. The platelet aggregation rate,platelet aggregation inhibition rate and cytochrome P4503A4 expression level in liver were tested by Western blot method. Results After treatment,the maximum platelet aggregation rate in experiment group were（ 37. 23 ±5. 01） % which significant lower than that of control group（ 57. 21 ±5. 23） %（ P〈0. 05） and the platelet aggregation inhibition rate in experiment group were（ 33. 56 ± 4. 64） % which significant higher than control group（ 21. 12 ± 3. 36） %（ P〈0. 05）. Compared with control and blank group,the liver cytochrome P4503A4 expression was significant decreased in experiment group（ P〈0. 05）. Conclusion The anti- platelet function for clopidogrel may caused by inhibition P4503A4 expression in rats.
