摘  要:目的探讨促红细胞生成素(EPO)对新生鼠缺氧缺血性脑损伤(HIBD)后海马颗粒下带(SGZ)5-溴-2-脱氧尿嘧啶(Brd U)表达的影响。方法选择7日龄新生Wistar大鼠制备新生鼠HIBD动物模型,按照体重将新生鼠随机分为假手术组、HIBD模型组、EPO实验组(EPO 5 U·g-1·d-1×14 d)。在术后第14天、第21天、第28天,动态检测海马SGZ区Brd U阳性细胞数。结果海马SGZ区Brd U阳性细胞表达:3组新生大鼠在术后第14至28天,Brd U阳性细胞数随着新生鼠日龄增加而明显减少(P<0.01);在术后第14天、第21天、第28天,3组新生大鼠的海马SGZ区Brd U阳性细胞数以EPO实验组最多,其次为HIBD模型组,假手术组最少。在术后第21天,假手术组、HIBD模型组、EPO实验组的海马SGZ区Brd U阳性细胞数分别为8.08±1.62,25.71±4.18,32.21±8.63。在术后第14天、第21天,3组间两两比较差异有统计学意义(P<0.01);术后第28天,实验组与模型组比较差异有统计学意义(P<0.05)。结论新生鼠HIBD早期给予EPO干预治疗对新生鼠缺氧缺血性脑损伤可能有神经保护作用。Objective To investigate the effect of erythropoietin( EPO)on the expression of 5- bromo- 2- deoxyuridine( Brd U) in subgranular zone( SGZ) of neonatal rats with hypoxic- ischemic brain damage( HIBD). Methods Animal HIBD model of seven- day- old newborn Wistar rat was made,and then the model rats were randomly divided into two groups: the model group of HIBD and the EPO trial group( EPO 5 U·g- 1·d- 1for 14 d). The expressions of Brd U in dentate gyrus were examined with immunohistochemical staining and image quantitative analysis in fourteen days,twenty- first days and twenty- eight days after the operation. Results The expression of Brd U in SGZ: The number of Brd U- positive cell in SGZ of hippocampal region was gradually decreasing accompany with the increasing age in three groups,from 14 days to 28 days after operation. The Brd U- positive cell were a significantly difference in the each groups( P〈0. 01). The number of Brd U- positive cell of the EPO trial group was maximum,the model group was second,the sham operation group was least. In fourteen days,twenty- first days after operation,the number of Brd U- positive cell in SGZ of hippocampal region of the sham operation group,the model groupand the EPO trial group were 8. 08 ± 1. 62,25. 71 ± 4. 18,32. 21 ± 8. 63,respectively. By pairwise comparison,there is a significant difference in each groups( P〈0. 01) in twenty- first days. In twenty- eight days after the operation,there was no significant difference between the model group and the sham operation group( P〈0. 05),but the significant differences were found between the model group of HIBD and the EPO trial group( P〈0. 05). Conclusion Exogenous EPO could increase the expression of Brd U in SGZ of hippocampal region of neonatal rats with HIBD,it suggest that EPO could promote the neurogenesis in subgranular zone of hippocampal region of neonatal rats with HIBD,it may be involved in the regeneration of neurocytes and play an important role of neuroprotectiv
