摘  要:目的制备载淫羊藿苷的聚乳酸一羟基乙酸共聚物(Pt。GA)纳米粒(淫羊藿苷纳米粒),分析其理化性质、体外淫羊藿苷的释放性能及对人成骨细胞的影响。方法采用超声乳化透析法制备淫羊藿苷纳米粒,通过透射电镜及粒度分析仪对淫羊藿苷纳米粒进行表征,体外释放实验检测纳米粒中淫羊藿苷的释放特性,噻唑蓝(MTT)法、碱性磷酸酶活性检测和钙结节染色研究淫羊藿苷纳米粒对人成骨细胞MG-63生长和分化的影响。结果淫羊藿苷纳米粒呈球形,平均粒径为(185.8±5.6)nm,具有良好的生物相容性及缓释性能;该纳米粒对人成骨细胞的生长无明显影响,但能明显促进成骨细胞分化,且呈浓度和时间依赖性。结论采用超声透析方法制备的淫羊藿苷纳米粒粒径均一,可缓释淫羊藿苷,具有良好的生物相容性及促进成骨分化作用。Objective To prepare icariin-loaded poly(lactic-co-glycolic acid) (PLGA) nanoparticles (icariin nanoparticles) and analyze their physical and chemical properties, the release of icariin, and the effects on human osteoblast cells in vitro. Methods Icariin nanoparticles were prepared by ultrasonic emulsification and dialysis method and characterized by transmission electron microscopy and particle size analyzer. The release studies in vitro were adopted to evaluate the release-control features. Thiazolyl blue tetrazolium bromide (MTT), alkaline phophatase activity assay, and calcium nodes dyeing were calried out to investigate the effects of ieariin nanoparticles on the growth and differentiation of human osteoblast cells MG-63. Results The ieariin nanoparticles had spherical shapes and uniform particle size, and the mean size of the nanoparticles was (185.8±5.6) nm. The ieariin nanoparticles showed the best property of sustained release and biocompatibility. Meanwhile, the icariin nanoparticles had no effect on the growth of MG-63 cells, but they promote the differentiation of osteoblast cells by a concentration- and time-dependent manner. Conclusions The icariin nanoparticles prepared by ultrasonic emulsification and dialysis method have a good effect on the differentiation of human osteoblast cells in vitro and are biocompatible.
