牛蒡子苷元对实验性胃溃疡的保护作用及其机制探讨

摘　　要：目的:观察牛蒡子苷元对多种实验性胃溃疡的保护作用,并对其初步作用机制进行探讨。方法:连续皮下给予牛蒡子苷元(0.03 mg/kg、0.1 mg/kg、0.3 mg/kg)和灌胃给予西咪替丁(30 mg/kg)7 d后,禁食24 h,灌胃无水乙醇(0.1 ml/10g)制备胃溃疡模型,1 h后处死测定胃溃疡指数;连续皮下给予牛蒡子苷元(0.03 mg/kg、0.1 mg/kg、0.3 mg/kg)和灌胃给予雷尼替丁(50 mg/kg)7 d后,禁食24 h,皮下给予利血平(0.2 ml/10g)制备胃溃疡模型,6 h后,取血分离血清测定TGF-β、IL-1β以及测定胃溃疡指数;连续皮下给予牛蒡子苷元(0.05 mg/kg、0.15 mg/kg、0.45 mg/kg)和奥美拉唑(4 mg/kg)7 d后,放入(21±1)℃的水中,6 h后解剖,取血测定血清中超氧化物歧化酶(SOD)和诱生型一氧化氮合酶(i NOS),测定溃疡指数;连续皮下给予牛蒡子苷元(0.05 mg/kg、0.15 mg/kg、0.45 mg/kg)10 d和雷尼替丁(30 mg/kg)后,处死,测定溃疡指数,测定胃黏膜匀浆中丙二醛(MDA)水平。结果:牛蒡子苷元(0.03 mg/kg、0.1 mg/kg、0.3 mg/kg)皮下给予可以显著降低无水乙醇型小鼠胃溃疡的溃疡指数,牛蒡子苷元(0.1 mg/kg和0.3 mg/kg)可以保护无水乙醇诱导的胃黏膜损伤;牛蒡子苷元(0.03 mg/kg、0.1 mg/kg、0.3 mg/kg)可以显著减少利血平型小鼠胃溃疡的溃疡指数;牛蒡子苷元(0.05 mg/kg、0.15 mg/kg、0.45 mg/kg)可以显著抑制束缚水浸应激性大鼠的溃疡形成,显著升高SOD和i NOS水平;牛蒡子苷元(0.05 mg/kg、0.15 mg/kg、0.45 mg/kg)明显促进醋酸型大鼠胃溃疡的愈合,并且降低胃黏膜匀浆中的MDA水平。结论:牛蒡子苷元皮下给予对多种实验性胃溃疡具有保护作用,作用机制可能与升高SOD和i NOS水平和减少MDA水平相关,为其申报临床用于治疗胃溃疡提供临床前的药理试验依据。Objective： To observe the protective effects of arctigenin against gastric ulcer. Methods： The experimental gastric ulcer model was induced by anhydrous ethanol and reserpine in mice,and bondage water immersion and acetic-acid in rat,which the ulcer index and ulcer inhibition were measured. Results： The ulcer index of gastric ulcer induced by anhydrous ethanol were decreased significantly,the ulcer inhibition of arctigenin（ 0. 03,0. 1 and 0. 3 mg / kg） was 55. 9%,42. 1% and 40. 0%,respectively,at the same time,the pathology suggested the arctigenin（ 0. 3 and 0. 1 mg / kg） can protect the gastric injury. Also the arctigenin（ 0. 03,0. 1 and 0. 3 mg / kg） significantly decreased the ulcer index induced by reserpine. All doses of arctigenin（ 0. 05,0. 15 and 0. 45 mg / kg） can inhibited the ulcer induced by bondage water immersion through elevated the SOD and i NOS lever in serum. The ulcer index and the MDA level induced by acetic-acid were inhibited by all doses of arctigenin（ 0. 05,0. 15 and 0. 45 mg / kg）. Conclusion： The results of the present study show that arctigenin displays antiulcer activity,which may be associated with its alteration of SOD,MDA and i NOS. The study suggested a potential therapeutic application for arctigenin in the treatment of gastric ulcers.
